ThesisItem Open Access Isolation, evaluation and characterization of disease suppressing Plant Growth Promoting Rhizobacteria (PGPR) of lentil rhizosphere(ANDUAT, kumarganj, Ayodhya, 2021-07-31) Ahmad, Niyaj; Kumar, Dr. AdeshA total number of 16 lentil rhizobacterial isolates were isolated at Nutrient Agar medium. Only twelve isoaltes showed antagonistic activity against Fusarium oxysporum f. sp. lentil. All the antagonistic isolates produced diffusible and volatile antifungal metabolites. The isolates 10 and 14 showed a maximum antagonism 72% and 68.6% respectively. All the antagonistic isolates showed PGPR activity such as phosphate solubilization, IAA production and ammonia production. On the basis of their antagonistic and PGPR traits out of nine only two isolates, 10 and 14 were found to be more effective which were selected for SDS-PAGE and 16S rDNA molecular analysis. Strain 10 and 14 identified as Enterobacter ludwigii and Enterobacter cloacae. The protein bands of variable size are present and both isolates have similar protein banding pattern. The PCR was run with primers namely, 16s Forward 5`GGATGAGCCHAGGCCTA3` and 16s Reverse Primer 5`CGGTGTGTACAAGGCCCGG3` PCR product was run on 1.5% agarose gel for appearance of banding patterns sequence of PCR product of 16S rDNA, the electropherogram and dendogram generated which showed genetic similarity between the isolate 10 (Enterobacter ludwigii) and 14 (Enterobacter cloacae).ThesisItem Open Access Studies on microbe mediated biochemical and molecular mechanism responsible for induced systemic resistance (ISR) and plant growth promotion in Lentil (Lens culinaris Medik.) under stress of pathogenic “Fusarium oxysporum f. sp. lentis(ANDUAT,Kumaraganj,Ayodhya, 2020-10-29) Srivastava, Shalini; Kumar, AdeshA total of 20 rhizospheric strains were isolated at NA, King’s B and Jension media. Out of 20, only eight isolates (40 percent) showed antagonistic activity against Fusarium oxysporum f.spp.lentis. All the antagonistic isolates produced diffusible and volatile antifungal metabolites. The isolates (B5) and (B8) showed a maximum antagonism as 77.8% and 70% respectively. All the antagonistic isolates showed PGPR activity such as phosphate solubilization, Zn solubilization, Siderophore production ,Ammonia production, HCN and IAA production. On the basis of their antagonistic and PGPR functionality, out of eight, only two isolates, B5 and B8 were found to be more effective which were selected for ISR against wilt of lentil and characterized B5 and B8 strains by 16SrRNA molecular primer. Strain B5 and B8 identified as Bacillus safensis and Bacillus licheniformis respectively. The effect of these two bio-agents (Bacillus safensis and Bacillus licheniformis spp.) against wilt phyto-pathogen (Fusarium oxysporum f.sp.lentis ) lentil (Lens culinaris Medik.) was determined. Two varieties of lentil as resistant (IPL-316) and susceptible (K-75) were taken for the experiment.ThesisItem Open Access Molecular and biochemical screening of rice genotypes against salinity and drought (dual) stresses(ANDUAT, Kumarganj, Ayodhya, 2021-04-16) Singh, Vineeta; Dwivedi, D.K.The present investigation entitled “Molecular and biochemical screening of rice genotypes against salinity and drought (dual) stresses” was under taken for screening of twenty four rice genotypes namely, Taramon, NEDU, IR91167-31-3-1- 33, IR91167-99-1-1-1-3, IR91171-66-3-2-1-3, IR92953-49-1-3, NUD3, NDRK5088, NUD2, NDR359, CSR30, IR 29, IR64, CSR13, IR28, SARJOO52, AYAAR, AMKER, FL478, NDRK2008, Nageena 22, NDR 1, Baranideep and Sushk Samrat. Objectives were to evaluate rice varieties for salt and drought stresses (dual stress) at seedling and reproductive stages, to identify dual tolerant rice genotypes and to pyramid salt and drought tolerant gene(s)/QTL(s) in elite rice genotype. Screening of rice genotypes at seedling stage was carried out in hydroponics system in lab condition Twenty four rice genotypes after 21 days salinization tolerant genotypes were CSR 30 and CSR 13. The susceptible score 7 exhibited by NDR359, IR29 and IR 28. For drought stress screening at vegetative and reproductive stage observations were recorded for seventeen plant characters viz., seedling vigour (cm), days to 50% flowering, plant height (cm), panicle bearing tillers per plant, panicle length number of spikelets per panicle, number of grains per panicle, spikelet fertility (%), test weight (g), panicle bearing tillers per plant, biological yield per plant (g), harvest index (%), relative water content (RWC), leaf rolling, drought recovery and grain yield per plant and biochemical screening for drought and salinity with measurement of the chlorophyll content, proline content, sodium oxide dismutase (SOD), Peroxidase, Catalase and total soluble sugar content were recorded. The analysis of variance involving 24 rice genotypes with regards to yield, yield components and quality traits was done for seventeen characters under drought and thirteen characters under normal conditions. The mean sum of square values for interaction effects of traits among 24 rice genotypes under drought treatment were found significant for almost all the traits except panicle bearing tillers and leaf rolling at reproductive stage.
