ThesisItem Open Access UNDERSTANDING THE ROLE OF BOVINE MILK EXOSOMAL miR-326 ON INFLAMMATORY SIGNALING IN Caco-2 CELLS(ICAR-NDRI, KARNAL, 2021) KUMAR, AMIT; SINGH, DHEERMilk is an excellent package of nutrition and thus it is an integral part of human diet. Recently, a new bioactive component of milk is being explored which can regulate the gene expression of the recipient i.e., milk exosomal miRNA. In our lab, the exosomal miRNAs of cow and buffalo milk have been profiled earlier and the functional validation is required. Previous studies have proved that miRNA-326 can regulate inflammatory signaling. Thus, the present study aimed to explore the role of bovine milk exosomal miR-326 on inflammatory signaling using human intestinal cell line i.e., Caco-2 cells. Firstly, the abundance of miR-326 was validated in cow and buffalo milk exosomes by qPCR, which was earlier been profiled in our lab. Results showed that the miR-326 is abundant in exosomes of cow and buffalo milk. Further, we identified the target inflammatory genes of miR-326 from literature which was later confirmed by Target Scan, miRANDA, picTAR, miRDB (online prediction tools) and also validated the target genes in vitro by qPCR in Caco-2 cells. To study the effect of miR-326 on inflammation, TLR4 (confirmed by Target Scan) and MYD88 (confirmed by miRANDA) genes were chosen as target genes. First, the Caco-2 cells were transfected with miR-326 mimic and then the expression of both the genes was analyzed in the transfected cells by qPCR. The expression of TLR4 was significantly down-regulated in the transfected cells and thus downregulating NF-kb signaling. Hence, the production of pro-inflammatory cytokines (TNFα, IL-1β, IL-6, IL-12) and type-1 interferons, which were required for the inflammatory response got inhibited. The cell proliferation after treating the cells with bovine milk exosomes was assessed and it was observed that the exosomes induced cell proliferation. Thus, this study concludes that the milk exosomes functionally deliver miR-326 inside the recipient cells which may regulate the inflammatory signaling in vitro.
