dc.description.abstractABSTRACT: Lead occurs naturally in the Earth‟s crust and is an environmental contaminant of increasing importance. It is a highly toxic heavy metal and affects all organs and systems of an organism causing several acute and chronic diseases. Naringin is a flavonone glycoside obtained from tomatoes, grape fruits and many other citrus fruits. It is a very common dietary constituent and is reported to have wide spectrum of pharmacological actions including significant vaso-relaxant effect on smooth muscles. In view of this, the present study is designed to know the modulatory effect of lead on the contractile or relaxant responses of various pharmacological agents on sheep tracheal smooth muscle in the presence of naringin in vitro. The control and lead acetate treated tracheal smooth muscle strips were exposed to the contractile and relaxing agents like acetylcholine, salbutamol, sildenafil and sodium nitro prusside both in the absence and presence of naringin in cumulative doses. At the end of each study the strips were preserved for histopathological studies. Mean EC50 value for contractile response of acetylcholine obtained in the absence of lead acetate was 1.99 X 10-7 M and in the presence of lead acetate it has decreased to 1.00 X 10-7 M with a significant left shift. Mean IC50 values for relaxant responses of salbutamol, sildenafil and sodium nitro prusside were 9.77 X 10-7 M, 1.69 X 10-6 M and 1.35 X 10-6 M respectively. In the presence of lead acetate there was a significant increase in the mean IC50 values of salbutamol, sildenafil and sodium nitro prusside to 2.75 X 10-5 M, 1.73 X10-5 M and 1.02 X 10-5 M respectively. In the presence of flavonone naringin, the mean IC50 values of relaxing agents salbutamol, sildenafil and sodium nitro prusside decreased to 9.33 X 10-6 M, 8.12 X 10-6 M and 9.12 X 10-6 M respectively.en_US
