Abstract
The present study was undertaken to evolve nucleic acid detection methodology that Would prevent aerosol contamination of the previous amplified product and non-specific amplification. The approach used in this study for achieving the above objectives were to incorporate dUTP in the place of dTTP and perform subsequent polymerase chain reaction (PCR) or loop mediated isothermal amplification (LAMP) reaction in the presence of enzyme Uracil DNA glycosylase and to use the lateral flow dipstick assay on the LAMP products. These two approaches are applied in the diagnosis of Leptospira and Mycobacterium avíum subsp paratuberculosis (MAP).
