dc.description.abstractThe present work involves the development of a novel method of contraception in male animals using a synthetic co-polymer of Styrene Maleic Anhydride (SMA). For the polymer preparation, a simple single pot RAFT polymerization technique was carried out. The 2-dodecyl-thiocarbonothioyl-thio-propanoic acid (PADTC) was synthesized in the first step which was then used as a catalyst to initiate the polymerization process between styrene and maleic anhydride. The prepared SMA was analyzed using NMR, FTIR, MALDITOF, Zeta potential analyzer for its physical properties which proved to be similar to the commercially available polymer. The in vitro viability and integrity of canine spermatozoa were tested using different concentration of polymer. The motility, acrosomal integrity, mitochondrial membrane potential and DNA integrity before and after incubation were tested and the results indicated that 2 mg dissolved in100 μl DMSOproduced necrozoospermia in 60 minutes. The micro structural alterations of the sperm were confirmed using Atomic force microscopy (AFM). The invivo studies were performed using bilateral injection of polymer hydrogel into the vas deferens of adult male Wistar rats (n=6). Those injected rats were cohabitated with female rats (1:2) post 15 days of surgery and proved to produce no litters. The results indicated that the SMA formed a chemical mesh inside vas deferens which was evident through Scanning Electron Microscope (SEM) and causes electrolytic interaction in the sperms passing through it and hence rendering it infertile for a conception. In conclusion, SMA would be a quantum leap in modern contraception when it comes to mass sterilization of stray dogs.en_US
