dc.description.abstractABSTRACT The present study entitled “Molecular Characterisation of an Ilarvirus infecting Apple” was conducted in the Department of Plant Pathology, Dr. Yashwant Singh Parmar University of Horticulture and Forestry, Nauni, Solan, Himachal Pradesh during the year 2017. Surveys revealed that the incidence of virus disease in different apple growing areas of district Shimla during 2017 ranged between 5 to 71 percent. The predominant symptoms prevalent in most of the cultivars observed were chlorotic, bright yellow discolouration of the leaves in the form of blotching, mottling, vein banding or yellowing, ringspots, line and oak leaf patterns. Twenty isolates of apple were collected from different localities. A detailed study of the symptomatology of ilarviruses in apple revealed the presence of mixed infection of apple mosaic virus (ApMV) and prunus necrotic ringspot virus (PNRSV) in most of the trees. These results were further confirmed through serological detection by using ALP based direct form of DAS-ELISA. However, the prevalence of PNRSV in ELISA tests was found to be very low in comparison to ApMV. Among all plant parts tested for ELISA, apple leaves were found to be the most suitable for the detection of Ilarviruses (ApMV and PNRSV). DAS-ELISA based indexing between April and August was found to be the most appropriate period for detecting the presence of ApMV and PNRSV in apple. RT-PCR based molecular detection also resulted in the successful detection of apple mosaic virus as CP gene of ApMV was amplified using total RNA isolated from apple using ApMV CP gene specific primers and an amplicon of ~614 bp was obtained. High incidence of infections calls for an evaluation of the health status of apple nurseries thereby emphasizing the need for developing a sound certification scheme for the production of virus free planting material of apple.en_US
