Abstract
The aim of this study to synthesis of Zn-chitosan nanomaterial and evaluate its
effect on stevia plant tissue culture for growth and stevioside and rebaudioside
contents. Nanosized (323.7 nm) mono-dispersed porous Zn-chitosan nanomaterials
have remarkable effect on stevia tissue culture. The highest % shoot induction
(64.2%), shoot length (2.13) was observed in (0.03%) Zn-chitosan nanomaterial
supplemented shoot induction media. Maximum numbers of shoots (4.23)/
microshoot was obtained at 0.01% Zn-chitosan nanomaterials on shoot multiplication
media. Highest rooting response (90.16%), root number (5.8) and root length (3.51)
was noticed at 0.03% Zn-chitosan nanomaterial augmented root induction media.
Randomly selected leaves of regenerated plantlets from three concentrations of Znchitosan
nanomaterials (0.01, 0.03 and 0.06%) were evaluated for stevioside and
rebaudioside content by HPLC. Results revealed that stevioside content (4.32 ) and
reaudioside-A content (1.30 ) was higher at 0.03% concentration of Zn-chitosan
nanomaterials in tissue culture raised plants compared to control (MS media).These
results indicated that Zn-chitosan nanomaterials could be an promising biological
active compound for further study in stevia for growth and stevioside and
rebaudioside content in vitro and in vivo.
