Abstract
The present investigation was carried out with twelve genotypes including
a check of custard apple in the Department of Plant Breeding and Genetics and
Molecualr Biology and Biotechnology, RCA, Udaipur. Isolated and purified
DNA was subjected to PCR based marker (RAPD) for assessment of genetic
diversity. The quality of DNA was determined by calculating ratio between A260
and A280. This ratio ranged from1.51 to1.87. The concentration of DNA ranges
between 3.74μg/μl (RCA076) to 3.10μg/μl (RCA081). Total 8 RAPD primers
out of 15 gave good amplified products with template DNA. They gave 32
scorable bands and 130 clear fragments. All 32 bands and fragments were
polymorphic with 4 polymorphic bands per primer. P-20 and C-04 primers were
the most informative in present investigation.
From RAPD profiling, Nei and Li and Jaccards matrix was obtained.
Similarity and dissimilarity range of Nei and Li coefficient varied from 0.00 to
0.86 and 0.14 to 1.00 and similarity and dissimilarity range of Jaccards
coefficient was 0.00 to 0.75 and 0.25 to 1.00 respectively. Maximum binary
squared Euclidean distance was 21 and minimum distance was 3. The
dendrogram was constructed by Wards method and clustering of different
genotypes was also done by Tocher’s method. Distribution of genotypes was
same in the both methods except two genotypes i.e. RCA111 and RCA053. The
genotype RCA053 had maximum genetic distance from the other genotypes.
