Abstract
Genetic divergence among 23 genotypes of mungbean (Vigna radiata L.) was assessed through morphological (10 phenotypic traits), biochemical (PER, EST and SOD isoenzyems) and molecular (RAPD, ISSR and SSR) markers. Significant genetic variance was observed for days to 50 % maturity, days to maturity, plant height, number of branches/plant(cm), number of pods/plant, pod length(cm), 1000 seeds weight(g), seed yield per plant (g), biological yield(g) and harvest index (%) amongst the morphological characters chosen for study, as influenced by three different environments during cultivations. Hierarchical Euclidean and Ward’s cluster analysis based on 10 morphological character tics grouped all the 23 genotypes into five and two divergent clusters respectively, under the three environments investigated. A total of 10 alleles were generated by three isozymes viz., peroxidase, esterase and superoxide dismutase. Isozymes of SOD and esterase each exhibited a maximum of four activity zones followed by peroxidase showing only two activity zones. Isozymes of esterase and superoxide dismutase showed 50 and 25 per cent of polymorphism with all alleles. Jaccard’s Similarity Coefficient values lay between 0.73 to 1.00 that divided all the genotypes into two main clusters. Genetic divergence and distance derived from isozyme analysis was found low due to the small number of polymorphic alleles. RAPD analysis showed 93.48 per cent of polymorphism. A total of 126 amplified bands were obtained that ranged between 200 bp to 2900 bp, using 15 RAPD primers. The 117 out of 126 bands were polymorphic. ISSR analysis showed 80 per cent of polymorphism. A total of 75 amplified bands were obtained that ranged between 200 bp to 2500 bp using 13 ISSR primers. The 60 out of 75 bands were polymorphic. SSR analysis showed high degree of polymorphism (86.66%). A total of 15 alleles were obtained that ranged between 100bp to 190 bp using 10 SSR primers. The 13 out of 15 alleles were polymorphic. The average PIC of RAPD, ISSR and SSR were found 0.32, 0.208 and 0.205 respectively.
