    GENETIC AND MOLECULAR ANALYSIS OF BROWN PLANT HOPPER RESISTANT GENE(S) IN RICE (Oryza sativa L.)

    The present research work was undertaken in the Department of Genetics and Plant Breeding, College of Agriculture, Raipur with the objectives of identifying and monitoring the functional resistance genes in Chhattisgarh, understanding the mode of inheritance of resistance to brown plant hopper in some selected resistant donors, knowing the allelic relationships among resistance genes present in selected donors and studying genetic diversity at molecular level in selected lines. The experimental materials consisted of (a) A set of 500 advanced breeding lines including susceptible checks and resistant checks, which were screened against brown plant hopper at Raipur during Kharif 2009. (b) The 100 selected genotypes including brown plant hopper resistant advanced breeding lines, susceptible checks and resistant checks, which were screened at Raipur during Kharif 2010 and 2011 to monitor the resistance. (c) F1, F2 and F3 populations of 9 crosses attempted for inheritance studies and F1 and F2 population of the 17 crosses attempted for allelic studies were screened against brown plant hopper to ascertain the genetic ratios. Screening was carried out in glass house following 0-9 scale of standard evaluation system of IRRI and plants with score 0-3 were classified as resistant and those with scores 5-9 as susceptible. (d) A total of thirty eight microsatellite (SSR) markers were used to assess the extent of genetic diversity across the twenty seven genotypes. Out of four hundred seventy seven advanced breeding lines seven lines viz., R1641-914-1-400-1, R1723-1411-1-355-1, R1630-1248-2-846-1, R1709-2320-1- 2445-1, R1711-2409-2-2492-1, R1723-1413-2-356-1 and R1473-521-249-1-1 showed highly resistant reaction and fifty lines showed resistant reaction over the years. The inheritance investigation uncovered that brown plant hopper resistance in R-RF-55, R1243-1224-578-1 and R1723-1413-3-357-1 was controlled by a single dominant gene, while a single recessive gene governed resistance in R1546-1328-1-90-1 and two independent dominant genes governed resistance in R1600-1124-2-618-1. The allelic studies revealed that gene for brown plant hopper resistance present in R1723-1413-3-357-1 was allelic to Bph1 (MTU15). The gene for resistance in R1519-773-3-581-1, R1677-3473-1-4301-1, R1470-347-136-1-1 and R1670-3267-1- 3920-1 was non-allelic to resistance gene bph5 (ARC10550) and bph7 (T12). The resistance gene present in R1519-773-3-581-1 was different from Bph1 (MTU15) and Bph6 (Swarnalata). Similarly resistance gene of R1677-3473-1-4301-1 was different from bph2 (H105) and Bph6 (Swarnalata). The twenty one SSR markers generated polymorphic patterns while seventeen SSR markers found monomorphic. A total of 61 alleles were detected in 27 rice genotypes and the number of alleles per locus ranged from 2 to 5 with an average of 2.9 per locus. Size differences between the smallest and largest alleles varied from 5 bp (approx) to 40 bp (approx). Polymorphism information content ranged from 0.14 to 0.72 with an average of 0.48. A dendogram based on UPGMA analysis showed the genetic diversity among the 27 rice genotypes and similarity coefficients ranged from 0.23 to 0.91. Analysis of clusters of dendogram revealed highest 91 per cent genetic similarity between genotypes Kariyabenikat K 1866 and Danwar2511 in cluster II and also between genotypes Danteshwari and Samleshwari in cluster I.

Summer and rainy season heterosis hybrids along with earliness and marketable colour index; parents IBWL, PPL and MK whereas, in case of hybrids PPL x WBPF, PPL x PPR, WBPF x PPR, PPC x PPR, IBWL x WBPF exhibited desirable performance of Chhattisgarh performance.

    .................................................................................................................The prfesent investigation was undertaken during kharif, 2003-04, 2004-05 and 2005-06 at Horticultural Farm IGKV Raipur to study the type of gene action involved for fruit yield and its components, to ascertain the magnitude of heterosis and inbreeding depression to determine the extent of variability, heriatibility and genetic advance for various quantitative characters, to estimate the general combining ability and specific combining ability of parents and hybrids, respectively and finally to study he association of fruit yield and its contributing traits in segregating generation (F2).
