SCREENING OF SORGHUM CULTIVARS DIFFERING IN YIELD FOR CARBON MONOXIDE (CO) SENSITIVITY OF CYTOCHROME C OXIDASE AND GA3 SENSITIVITY

dc.description.abstractThe present investigation was undertaken to monitor the redox state of cytochrome a. during in situ respiration in different sorghum cultlvars by carbon monoxide sensitivity of cytochrome c oxidase. The GA3 sensitivity, carbohydrate status of leaves and their correlation with grain yield of these cultivars were also studied. Seedlings of sorghum plants were grown in soil filled earthern pots under natural day-light condition at the Department of Biochemistry, Mahatma Phule Krishi 1991-92 Vidyapeeth, Rahuri during^for screening of sorghum cultivars for CO- sensitivity of cytochrome c oxidase. After germination the seedlings were daily irrigated with 15mM xii potassium nitrate. The leaves of 12 day-old seedlings were used to screen carbon monoxide sensitivity by measuring nitrite production during ±n vivo nitrate reduction assay under aerobic and anaerobic conditions. In CO aerobic assay of nitrate reductase after one min. exposure to CO, some cultivars of sorghum plants were CO-sensitive, whereas some cultivars were CO insensitive. However, during the subsequent incubation,the exact C0:02 ratio was not maintained. By exposing the leaf segments to precisely defined COx02 ratios, it was observed that all the sorghum cultivars were CO-sensitive at the COi02 ratio of 10 and 5, It indicates that the cytochrome a^ in these cultivars was largely in the reduced state. These cultivars were tested for ibberellin (GAO and thesensitivity^ it was found that all^ested cultivars were insensitive to GA^. It was observed that there was no significant correlation between carbohydrate status of leaves and yield with carbon monoxide sensitivity.en_US
