Abstract
Experiment was carried out in three stages, stage one was named as experiment 1 in which wheat seeds of var HUW-468 were soaked for 12 hours in four different concentrations (ranging from 2.0 to 10 mM )of Boric acid (H3BO3), and then dehydrated under forced air to find out the effects of Boron priming on germination. Treatments are (T0)- control non primed seeds, (T1)- seeds primed with 2mM boric acid, (T2)- seeds primed with 4mM boric acid, (T3)- seeds primed with 8mM boric acid,( T4)- seeds primed with 10mM boric acid. Each treatment has four replications. Various parameters like germination percentage, plumule , radicle and seedling lengths, fresh and dry weights of radicle and plumule, alpha amylase activity, soluble sugar contents were determined at 3, 5, 7 and 9th days during germination and seedling growth and compared with non- primed control. Experiment 1 was performed to get the best boron concentration and after that Experiment 2(A) was conducted in which the same above parameters were studied by taking treatments like (C0)-non primed seeds (control), (C1)- seeds primed withMg(NO3)2 (7.5 mM), (C2)- seeds primed with H3BO3 (8mM), (C3)- seeds primed with H3BO3& Mg(NO3)2, (C4)- seeds primed with distilled water (hydroprimed). Experiment 2(B) was carried out by taking leaf samples from 30, 45, 60th days old plants planted in pots and data was collected based on biochemical parameters like chlorophyll, proline, protein & SOD contents and nitrate reductase activity along with SDS PAGE protein profiling. The work has been discussed on the basis of priming effects of magnesium nitrate and boron either used alone or in combination to wheat variety HUW-468.
