Abstract
The present investigation was aimed to develop a technique for the high frequency of regeneration of clonal apple rootstock mm106 using leaves as explants of in vitro shoot cultures. MS medium was found suitable for regenerating of mm 106 as compared to the LS medium. adventitious shoot formation was observed on 3 out of 8 combinations of BAP while direct organogenesis was obtained on 2 combinations out of 3 the highest regeneration frequency (37 % ) was observed ub MS medium supplemented with 22 um BA and 5.4 um NAA with 6.5 shoots per explant was pbtained in 8.8 um BA and 5.4 um NAA shoot number was found to be increased with the increase in induction period. higher potential for regeneration was observed when first two apical and expanded leaves form 30 days old cultured were used as explants. but the apical leaves must be of sixe of 5-8mm repeated regeneration cycle showed that the regeneration percentage decreases as the number of cycles increases. regenerated shoots of more than 2 cm were utilized for rooting. 60% rooting was obtained. the rooted plantets wee transferred to plastic posts having per sterilized potting m9izture soil, and and compst (1:1:1). 70% survival rat4e was obtained in pots .
