dc.description.abstract | Leptospirosis is a highly endemic disease in Kerala affecting man and animals. Microscopic Agglutination Test (MAT), though, the gold standard test for leptospirosis is often a less preferred choice since it is very cumbersome and serovar specific. Alternatively, genus specific tests like ELISA are considered more appropriate for routine diagnosis of leptospirosis. In the present study, recombinant outer membrane proteins of Leptospira,viz. rLipL21 and rLipL32 were used as antigens in ELISA for detection of IgG specific to Leptospira. The corresponding genesie., lipl21 and lipl32 were cloned,expressed and purified to produce recombinant proteins. Sera samples collected from 124 suspected cases of leptospirosis were subjected to MAT and were subsequently tested in ELISAs. The sensitivity and specificity of rLipL21 ELISA was found to be 90.90% and 73.52%, respectively. Similarly for rLipL32 ELISA the values were 81.81% and78.43%, respectively. On statistical analysis it was observed that both the ELISAs were promising tests for the diagnosis of leptospirosis as compared to MAT.
