dc.description.abstractSesame (Sesamum indicum L.) is an important oilseed belongs to family Pedaliaceae. In the present study, trait (yield and mineral) associated microsatellite markers were identified and validated in sesame genome. Identified marker were utilized in diversity study and cross-species transferability. For the transferability analysis 3 genotypes of S. malabaricum and 2 genotypes of S. prostratum were used. Total 50 microsatellite motifs were identified from genomic regions (LG 8, LG 11 and LG 16) of sesame. Across the identified markers, di-nucleotide repeats were abundant (58%) compare to other motifs. Total 18 primers were synthesized and validated in S. indicum. Out of 18, 12 were amplified with accepted amplicon size. For diversity and transferability analysis 11 additional primers from published literature were also taken. Out of 11 randomly selected primers, only 4 primers were amplified. In cross-species transferability analysis, S. malabaricum and S. prostratum showed transferability of 75% and 81.25%, respectively. Out of amplified 16 primers, 4 primers showed polymorphic appearance and 5 primers shows monomorphic appearance and other primers not completely amplified. Amplification in wild relatives indicates that developed markers can be used in other than S. indicum species. For diversity analysis, 7 primers (58.33%) showed polymorphic appearance. All polymorphic primers were trait associated. Polymorphic information content (PIC), major allele frequency and gene diversity of markers ranged from 0.275 to 0.528 (mean: 0.396), 0.545 to 0.792 (mean: 0.649) and 0.330 to 0.595 (mean: 0.470), respectively. These trait specific markers can be utilized for the marker assisted breeding, diversity analysis and wild hybridization with wild relatives of sesame.en_US
