Abstract
The present investigation “Electrophoretic profile of seminal plasma proteins vis-à-vis in-vitro sperm characters in Ongole bulls (Bos indicus)” was undertaken to evaluate the physico-morphological characteristics, to assess the in-vitro sperm characters and to analyze the electrophoretic profile of seminal plasma proteins in Ongole bulls. In the present study, the mean ejaculate volume (ml), mass activity (0-4), individual motility (%), sperm concentration (x106/ml), percent live spermatozoa, sperm abnormalities (%), acrosomal integrity (%) and HOST (%) in fresh semen were recorded to be 3.97 ± 0.25, 3.24 ± 0.11, 75.14 ±0.82, 1172.58 ± 57.19, 76.52 ± 0.56, 78.35 ± 0.68 and 57.85 ± 0.84, respectively. Individual variation between bulls in various semen characteristics such as ejaculate volume, sperm concentration, live sperm count and acrosomal integrity were observed whereas other parameters such as mass activity, initial motility, sperm abnormalities and HOST did not vary between bulls. None of the fresh semen characteristics varied between the ejaculates. Physico-morphological characteristics of frozen semen revealed 52.12±1.03 percent post thaw motility, 63.83±1.78 % live sperm count, 11.32% sperm abnormalities, 68.07±1.59% acrosomal integrity, 32.23±1.33% HOST and 22.65±0.81 mm CMPT. In-vitro fertility parameters (HOST and CMPT) did not show variation between bulls. The overall mean fertility rate was found to be 66.58±2.79 percent with a range of 59.48 to 79.41 percent with no significant difference between bulls. The total protein concentration of the seminal plasma was found to be 67.47±6.10 mg/ml. One dimensional SDS-PAGE of seminal plasma revealed as many as seven protein bands ranging from 22 to 170 kDa. The observed protein bands are between 22-29 kDa, 30 kDa, 41 kDa, 42-52 kDa, 53-70 kDa, 93-129 kDa and 130-170 kDa. A protein band with 41 kDa molecular weight was prominent in one of the breeding bulls which had highest fertility rate of 79.41%. It was concluded that individual variation among bulls was observed in respect of ejaculate volume, sperm concentration, live sperm percent and acrosomal integrity of neat semen. Post thaw sperm membrane integrity (HOST) and cervical mucus penetration distance (CMPT) by spermatozoa did not vary between bulls. Post thaw sperm characteristics indicated that all the bulls of the present study have been donating semen which could sufficiently withstand to freezing damage.
