Abstract
The present investigation entitled Phenotyping and molecular marker analysis of different population derived from cross DBW17 x WH1105 of wheat , consisted of parents (DBW17 and WH1105), F3, BC1F2 and BC2F2 population to study the inheritance and molecular aspects of yellow rust resistance and genetics of quantitative traits including grain yield and its component traits. The characterization was done on the basis of 11 morpho-physiological traits. F3, BC1F2 and BC2F2 populations had ample amount of variability for all traits under study. These traits can be used for selection in further generations. The correlation studies revealed the positive relation of all the traits with grain yield except days to heading and plant height in all the three populations. On the basis of cluster analysis 100 plants of F3 generation were grouped into seven clusters while 100 plants of BC1F2 were grouped into 9 clusters and 50 plants of BC2F2 were grouped into 7 clusters based on the relative magnitude of their D2 values. Molecular analysis was also done using 99 SSR markers, 15 were found polymorphic among parents and their three populations. It was confirmed by NTSYS-PC UPGMA cluster tree and dendrogram that two parents were diverse genetically and the progeny lines were interspersed between the two parents in F3 population and in both backcrosses progeny lines resembled their respective recurrent parents. Two and three dimension principal component analysis (PCA) based on SSR markers data showed similar clustering as was evident from cluster tree analysis. Single marker analysis of primers revealed that primers on nine chromosomes were associated with grain yield per plant and other yield attributes in all the three populations. The marker Xgwm374 was most tightly linked with the spike length at 0.01% significance. In BC2F2 population marker Xgwm582 showed linkage with yellow rust locus and other yield attributes at 5% significance. These markers can be used for selection and pyramiding of the genes.
