dc.description.abstractABSTRACT : A study to investigate the prevalence of Theileria infected Hyalomma ticks of crossbred cattle were carried out in arid to semi arid regions of Rayalaseema region in Andhra Pradesh using methyl green pyronin staining technique (MGP) and polymerase chain reaction (PCR). Hyalomma ticks were collected during March to July, 2014 from healthy and pyrexia noticed crossbred cattle irrespective of age, sex, and weight. The examination of 307 semi-engorged female Hyalomma ticks by genus and species (T. annulata) specific PCR using primers derived from gene encoding ssrRNA and Tams1 (T. annulata 30 kDa major merozoite surface antigen) revealed positivity of 16.94% and 5.21% in Hyalomma ticks. Per cent prevalence of Theileria was reported to be more in H. margiantum (18.18%) in comparison to H. anatolicum ticks (15.0%). Further, 4.17% of T. annulata prevalence was reported from H. a. anatolicum ticks. Since the MGP staining revealed positivity of only 3.90%, the PCR assay could be the optimum tool in the diagnosis of Theileria infection in Hyalomma ticks. No theilerial organisms were detected in Boophilus microplus and Rhipicephalus haemaphysaloides. Finally, the sequence analysis confirmed the 99% homology of Tams1 gene with tH1 strain of T. annulata.en_US
