EFFECT OF BIOFLOC BASED CULTURE SYSTEMS ON MICROBIAL COMPOSITION, GROWTH AND IMMUNE RESPONSE IN PACIFIC WHITE SHRIMP, LITOPENAEUS VANNAMEI

Abstract
A 49 day indoor trail was conducted with three treatments and one control to
evaluate only biofloc (T1), biofloc+substrate (T2) and only substrate (T3) based
rearing systems on the microbial composition of rearing water, growth performance
and immune response of pacific white shrimp Litopenaeus vannamei. Wheat flour
was used as carbon source to maintain C/N ratio at 15:1 in treatments T1 and T2.
Each experimental group consisted of four replicates in rectangular HDPE tanks of
70 lit capacity filled with 10 ppt salinity treated sea water were stocked with 10
numbers/tank of uniform sized L. vannamei juveniles (4.56 ±0.13 g). The net
weight gain, SGR, FCR, FER and survival rate were significantly higher (P < 0.05)
in the treatment groups than control. Among the treatments, higher net weight gain
(10.38 ± 0.14 g), SGR (2.42 ± 0.02), FER (8.85 ± 0.09), survival (77.50 ± 0.95 %)
and lowest FCR (1.27 ± 0.12) were recorded in T2. The water quality parameters
viz., DO, water temperature, pH, salinity, total ammonia-N (TAN), nitrite and nitrate
were in manageable range for L. vannamei. The addition of wheat flour in the
treatments T1, substrate in T3 and both wheat flour and substrate in T2 has
significantly reduced TAN, nitrite and nitrate in treatment tanks than the control. At
the end of the experiment, water samples analyzed for microbial population has
shown significantly higher (P < 0.05) total heterotrophic bacteria (THB), Bacillus,
Lactobacillus and Vibrio counts in all the treatments than control. Among the
treatments, significantly higher microbial count was recorded in T2 followed by T1
and T3. The haemolymph and serum samples collected at the end of the
experiment to study the immune status of shrimp has shown significantly higher (P
< 0.05) total haemocyte count (THC), total hyaline cells, total granuloc ytes, serum
protein, phenoloxidase (PO) activity, serum lysozyme activity in treatment groups
when compared to the control. Among the treatments, T2 with biofloc and substrate
combination has recorded with significantly higher THC (47.24 ± 4.49 x 106 cells ml-
1), hyaline cells (18.21 ± 2.22 x 106 cells ml-1), total granulocytes (29.04 ± 2.89 x 106
cells ml-1), serum protein (82.67 ± 0.01 mg/ml), serum PO activity (0.73 ± 0.03 OD
at 490 nm) and serum lysozyme activity (56.32 ± 0.03 %). The study revealed that
biofloc, substrate and biofloc+substrate combination systems have performed
significantly better in terms of water quality, beneficial bacterial loads in water,
growth performance and immune response in L. vannamei juveniles. Further, it
could be concluded that the integration of substrates in BFT has resulted in higher
production of L. vannamei.
