Abstract
The investigations were carried out to understand the genetical variations amongst the isolates of SclerotiumrolfsiiSacc. inciting collar rot of chickpea. Thirty six isolates from different chickpea growing areas of North zone, North –West plain zone, North-East plain zone, central zone and southern zone of India were isolated. The pathogenicity of all the isolates was confirmed by using chickpea variety JG-62. All the isolates were found to be pathogenic exhibiting 100% mortality. The variations in latent period ranged from 62 hrs to 122 hrs in different isolates. In the isolate CZCHRAI it was minimum, while in the isolate NEPZBMOK it was maximum. The morphological characters of mycelium and sclerotia of S. rolfsii isolates were studied using PDA medium. Milky white colony color was observed in 20 isolates, 12 isolates were white and 4 isolates were dull white, which indicated color differences in various isolates. The appearance of colony in isolates was found to be varied. Out of 36 isolates 19 isolates showed dense cottony growth, 12 were loose cottony and in 5 isolates growth was sparsely cottony. In 25 isolates mycelial threads were thick and in 17 isolates mycelial threads were thin. The variations in sclerotial morphology amongst the isolates were observed. The sclerotia formed per Petri plate varied from 58 in the isolate CZMSRHR to 990 in the isolate CZCHRAI. Their arrangement in Petri plate also varied. In most of the isolates sclerotia were found to be spread all over the plate. In the isolates NZAMSHI, NWPZIARI, CZMSDHU, CZGUJAND and SZKTRAI sclerotia were formed in circular form at the periphery of the Petri plate.The isolate CZGUJN produced sclerotia at the centre as well as at periphery of the plate. Variations in sclerotial size were noted in different isolates. Seventeen isolates bear small sclerotia, 10 isolates had medium sized sclerotia and in 9 isolates they were bold. In the isolate NWPZIARI the test weight of 100 sclerotia was 372.50 mg which was maximum as compared to the other isolates and minimum in the isolate NEPZBMOK having 23.00 mg of 100 per sclerotial weight.
