    Studies on certain gastro-intestinal nematodes with special reference to those found in goats

    Life history of Triohuris globuloss has been elucidated for the fist time using kids !q as experimental animals. Both free living and parasitic stages have been described in detail with illustrations. The prepatent period of Triohuric globuloss was found to be 85-89 days. T.globulosa failed to develop in guniea pig on experimental transmission. Haematology and histopathology of trichuriacis in kids experimentally infected with T.globulosa have been studied for the first time and heavy infection with the worm was found to cause anaemia and pathology changes in cacum of the host. Prepatent period of Strongyloides napillosus in kids infected orally with 7000 larva has been determined for the first time to be 6-7 days. Neoasoais vitulorum of calves has been found to be not transferable to oat either transplacentally or orally. In gunia pigs M.Vitulorum failed to attain sexual maturity. Calves could not be infected with infected with M.vitulorum post-nataly. Oesphagostomum colubianum of goats has been found to be not transferable to calves. Prepatent period of O.colusbionum in kids infected experimentally was found to be 40 days. Comparative efficacy of 4 anthelminties viz., Oxibendazole, thiophanate, Albendazole and Parbendazole was increased by conducting treatment trials. Against monospecific infection of Trichuris globulosa in experimentally infected kids the efficacies of Oxibendazile, thiophanate, Albendazole and Parbendazole at the recommended doses of 10mg.50mg, 10mg and 30 mg/kg body weight respectively were found to be 48.6%, 41%,47.3% and 49.3% respectively. Even at double the recommended doses the anthelminations were found to be well tolerated by the kids. Oxibendazole and Albendazole at the rate of 10mg and 10mg/kg body weight respectively were found to be 100% found to be 100 % effective against colubriformise, effective against respectively were Haemonohus contortue, trichostrongylus .Thiopanate at the recommended dosage (59mg/kg body weight) was also 100% effective against all he nematodes mentioned above except Haemonous contortus against which it was only 97% effective at that dosage. Parbendazole at the recommended dosage shedule (30mg/kg body weight) was 100% effective against the nematodes with the exception of .against which only 86.6% efficacy was noticed. Against in nature nematodes in the gastro-intestinal tract Oxibendazole and Albendazole were 100% effective whereas the efficacies of P`arbendazole and thiophanate were 85% and 74% respectively at their recommended doses. In in vitro studies the anthelmintic efficacy of Oxibendasole against Haemonchus contortus was found to be superior to that of the remaining anthelminties. In ovicidal property also, Oxibendazole was better than all the others, since it was ovicidal in 6 hours after administration whereas albendazole, thiophanate and Parbendazole were ovicidal in 8 hours, 9 hour and 11 hours respectively. Albendazole was found to be superior to al the other 3 anthelmintion in their influences on the body weight gain of treated animals closely followed by Oxibendazole. Perbendazole and thiophanate were inferior to others Judging on the basis of over all efficacies Oxibendazole and Albendazole were found to be superior to thiophanate and Parbendasole. Treatment trials with Oxibendazole, Albendazole and thiophanate against gastro-intestinal nematodes of goats have not been attempted earlier than the present study.

    Postnatal development of testis and epididymis, semen characteristics and fertility of brown- swiss crossbred bulls

    A systematic investigation involving the use of 24 Brown Swiss crossbred bull calves and 7 adult bulls with 62.5% exotic inheritance varying in age from 0- 360 days and 22-40 months respectively, resred and maintained at Indo-Swiss Project, Mattupatty, Kerala, was carried out in order to (a) trace the postnatal growth and development of the testis and epididymis and (b) assess the semen characteristics such as volume, initial motility, sperm concentration, live sperm count and incidence of abnormal sperms; As a corrolary to and as an integral part of this comprehensive study, 6679 semen collection particulars from the records maintained at the Indo-Swiss Project, Mattupatty and 7586 insemination data at the Crossbreeding Research Centre, Muvathuphza werecritically scrutinised to deduce freezability/discard rate of the ejaculates and the fertility potenoy of the bulls, respectively. The materials used, the salient observations made and the valid inferences drawn aare given below: Twenty four pairs of testis and epididymis collected from the bull calves were processed to study the postnatal growth and development. The pattern of growth of testis and of the epididymis was found to be curvilinear. The testicularand epididymal weights were observed to be significantly (p<0.01) correlated with the age, and body weight of the animals. A highly significant (p<0.01) correlation between epididymal weight and testicular weight was also observed. The growth rate of the seminiferous tubules appeared to be of sigmoid type. Formation of lumen in the seminiferous cords was initiated at 150 days and completed, at 180 days of age. The transformation of gonocytes to spermatogonia occurred at 150 days of age. The formation of primary spermatocytes, spermatid and spermatozoa cocurred at the age of 150 days, 300 days and 300-360 days, respectively. The process of transformation of supporting cells was initiated at 120 days and completed at 180 days of age. The transformation of immature Leydig cells to mature cells was initiated at 150 days and completed, by 180 days of age. There were distinct regional differences between the caput, the corpus and the cauda epididymis in respect of the diameter of the tubules, height of the epithelium and the degree of transformation of Epithelial lining from simple columnar to pseudostratified type. The distal part of the ductus epididymis seemed to have attained the adult size at an earlier age than the proximal. Sperms were seen in all the three regions at about 300 days. The mean values of ejaculate volume, initial motility, sperm concentration, live sperm count, sperm head abnormalities, tail abnormalities and proximal protoplasmic droplets were found to be 2.65 ± 0.84 ml, 66.24 ± 0.87%, 1599.64 ± 55.371 milions per ml of semen, 72.55 ± 1.310%, 4.91 ± 0.439%, 0.65 ± 0.045%, 6.47 ± 0.775% and 1.20 ± 0.143%, respectively. Significant (p<0.01) variations between bulls were observed in respect of sperm head abnormalities, tail abnormalities and proximal protoplasmic droplets and between months, in respect of ejaculate volume, sperm head abnormalities, free loose head, tail abnormalities and proximal protoplasmic droplets. Seasonal variation (p<0.05), however, was evident only in the incidence of tail abnormalities. Variations due to age of bulls were found to be significant (p<0.01) in the casse of ejaculate volume, sperm concentration, free loose head, middle piece abnormalities and tail abnormalities. The freezability of sperms expressed in terms of per cent of post-thawing motility was found to be 36.86. There was no seasonal effect on the freezability of sperms. The freezability rate increased significantly (p<0.01) with advancement of age of bulls. The overall rejection rate of the ejaculates was noted to be 39.80%. Most of the rejection (71.75%) was done before freezing. The pre-freezing rejection was found to be 28.55% of the total ejaculate. The post-freezing discard to be 15.74% of the frozen samples. The rejection rates of the ejaculates differed significantly (p<0.01) between months/seasons/age of the bulls. The rate of rejection was found to decrease with advancing age. The overall fertility rate of the bulls was found to be 56.06%. There were significant (p<0.01) differences in the fertility rate between months/seasons of insemination and between age of the bulls. As between bulls also, there was a significant variation (p<0.01) in the fertility rate. The significance of the observations inferences indicated above and their relevance to the implementation of cross breeding programme in Kerala were discussed briefly.

    Studies on the common ticks affecting live-stock in Kerala

    An investigation was conducted for a period of 5 years from 1974 to 1979 in to the incidence of ticks on domestic animals in Kerala, the biology of the most common species, and their susceptibility to four commonly available organo phosphorus compounds.. Nine different species of ticks belonging to 5 genera were collected from cattle, buffaloes, goats and dogs in different localities of the eleven districts of Kerala. The species of ticks identified were boophilus annulatus, rhipicephalus sanguineus, R.haemaphusaloides, R.turanicus, haemaphysalis bispinosa, H. turturis, H.spinigera, Hyalomma anatolicum and Amblyomma integrum. The commonest among them was boophilus annulatus, which was found mainly on cattle. The incidence of this tick was very high in the hilly areas and in the western ghat regions. The other common ticks found on cattle were Rhipicephalus haemaphysaloides and Haemaphysalis bispinosa. Tick infestation was found to be rare in buffaloes and goats and the common species found on them were Haemaphysalis bispinosa and B. annulatus. IN dogs the species recorded in the present study were Rhipicephalus sanguienus and R. turanicus. Variations in incidence was evidenced from the present study. The maximum incidence was found in the months of July, August, September, October and November. The life-cycle of B.annulatus was completed experimentally in a period 44 days. The maximum number of eggs produced by one tick was 1420 and minimum 680. The incubation period was dependent on the room temperature and variation in humidity had little influence. The larvae were found to prefer hairy areas of the body for attachment, and they did not attach at the hairless and exposed areas. The engorged females were found to drop off quickly when green grass was spread on the floor of the experimental animal’s shed. The life-cycle patter could be summarised as: Pre-oviposition : 1 to 6 days Oviposition : 5 to 9 days Incubation period : 16 to 30 days Larval fasting : 2 to 5 days Larval feeding : 4 to 7 days Larval moulting : 8th to 12th day of infection Nymphal feeding : 3 to 6 days Moulting of Nymph : 15 to 19th day of infection Adult feeding : 5 to 10 days Detatchment of female : From 20th day of infection onwards Four organophosphorous compounds viz. Malathion, Fenitrothion, Dichlorvos and Phosalone were tested for their action on ticks. The ovicidal and larvicidal actions and the lethal effects on nymphas and engorged females of B. annulatus, R sanguinius and H. bispinosa were studied in the laboratory. Cythion (malathion 50%) 0.5 to 1%, Sumithion (Fenitrothion 50 %) 0.5 to 1%, Dichlorvos (Nuvan) 0.1 to 0.2%, and Zolone (Phosalone 35 %) 0.2 to 0.3% were found effective at different degrees. The efficacy was directly proportional to the concentration of compound and the duration of treatment. Phosalone showed maximum efficacy followed by Dichlorvos. In field trials, all the four compounds gave satisfactory results. But phosalone showed maximum efficacy and least toxicity to animals. Nuvan was also effective but was the most toxic among the four compounds. Thus the acaricide of choice for routine use in Kerala was determined to be Phosalone (35%) at a concentration of 0.3% to be used as a spray or wash.

    Pathology of the reproductive organs in experimental hypothyroidism in goats

    A survey study was conducted to assess the role of hypothyroidism in the etiology of reproductive disorders in goats. Lowered functional activity of the thyroid was associated with postpartum anoestrum, repeat breeding and delayed puberty. An experimental model of controlled hypothyroid state was induced in goats, using different dose regimes of thiourea with the objective of studying the sequence of physiopathological changes in hypothyroidism and its influence on reproduction. Clinically healthy young and adult cross-bred goats of both sexes were employed for the study. The animals were divided into control, Treatment I (T1) and Treatment II (T2) groups. Groups T1 and T2 were administered thiourea orally at the rate of 50 mg/kg and 100 mg/kg body weight respectively. Clinical symptoms and growth rate were studied. Protein bound iodine, total serum protein and serum cholesterol were estimated. Haematological data were collected and the semen characteristics were evaluated. On the 90th day of the experiment both control and experimental animals were sacrificed leaving at least one animal in each group. These animals were maintained without treatment with thiourea for a further period of 90 days and sacrificed. Cross lesions and histopathological changes in tissues were recorded. Weakness, lethargy, depression, reduced feed intake, subcutaneous oedema of varying degree, loss of libido in males and anoestrum in females were the important clinical features observed. There was stunting of growth and apparent reduction in weight of the animals dosed with thiourea. On discontinuation of treatment with thiourea all the clinical signs manifested disappeared and the animals gained weight progressively. There was significantly decrease in the protein bound iodine, and increase in the total serum protein and cholesterol level in all the animals dosed with thiourea. The values reached the normal level on discontinuation of treatment with thiourea. Microcytic hypochronic anaemia was evident in hypothyroid goats. There was improvement in haemogram values on withdrawal of thiourea administration. Reduction in quantity and quality of semen was observed in hypothyroid goats. The semen appeared normal in quality and quantity on discontinuation of thiourea administration. Gelatinisation of subcutaneous fat and hypertrophy and dilation of the left verntricle were the common findings at autopsy. There was significant increase in the relative weight of the thyroid, pituitary and the adrenal gland. Histologically the thyroid glands exhibited varying degree of hyperplasia and hypertrophy. Predominant histological changes in the pituitary was hyperplasia and hypertrophy of basophil cells and degenerative changes in the acidophils. Adrenal gland showed hypertrophy and depletion of fat in the zona fasciculata. On discontinuation of treatment with thiourea all the lesions disappeared. A significant decrease in the relative weight of the testis and epididymis was observed in all the goats dosed with thiourea. Histologically the seminiferous tubules were small and lined by single layer of spermatogonial cells. Some tubules contained degenerated and desquamated cells without any evidence of sperms. The tubules of the epididymis were small in size and the lining cells showed deciliation and desquamation. These degenerative changes were found to be reversible on discontinuation of treatment with thiourea. The accessory sexual glands, the seminal vesioles and the prostate weighed less in hypothyroid goats. Histologically the cells lining the glands were small inactive, vaculated and hyalinised. After discontinuation of administration of thiourea the reparative changes were almost normal. There was significant decrease in the relative weight of the ovary and uterus in all the animals dosed with thiourea. Histologically ovaries were found to be inactive and only few scattered immature follicles were seen. Uterine glands were few and non secretary type. The lining epithelial cells did not show active mitosis. On discontinuation of treatment with thiourea the weight of the ovary and uterus increased and histologically ovaries and uterus were found to be normal. Kids and female goats were more susceptible to the effects of hypothyroidism. The significant pathological changes observed in the reproductive organs have clarified the importance of hypothyroidism in inducing sub-fertility and infertility in goats. The reversible nature of the lesion was also proved.

    Ochratoxicosis in the goat

    An experimental study was carried out to delineate the pathological effects of ochratoxin in goats. A comparative assessment of ochratoxin production by A. ochraceus and A. sulphureus on what and rice under static and shake cultures was also made. A. ochraceus was found to be a better toxin producing strain in both substrates under static and shake cultures systems and wheat was a better substrate than rice. Toxicity studies were conducted in Sannen – Malabari cross-bred goats of 1 to 3 months age. Purified ochratoxin produced in the laboratory was administered by oral, intra-peritoneal and intravenous routes. The different dose levels adopted were 2.5 mg/kg body weight, 1 mg/kg body weight and 0.5 mg/kg body weight. The synergistic effect of ochratoxin and aflatoxin in goats was studied by adminstering the crystalline toxins simultaneously (Makor Chemicals, Israel) by itraperitoneal route. The parameters of study were: clinical signs, haematological and biochemical alterations, pathological changes in urine, and macroscopic, microscopic and ultra-structural alterations in organs. Varying degree of clinocopathological changes were noticed in the test animals. The animals became weak and listless and in general there was reduction of total erythrocyte count, PCV, haemoglobin and lymphocyte count. Serum protein level was lowered while BUN and creatinine and blood coagulation time were high. There was rise in ALP, SGOT and SGPT in some of the test animals. The changes and degree of variation depended on the dose, total quantity and rate of administration of the toxin and duration of the experiment. More severe alterations were noticed when ochratoxin and aflatoxin were administered simultaneously. Important changes in the urine were lowering of pH, albuminurea and presence of epithelial cells and casts. Pathological changes varied in severity in different organs and were observed in the following descending order: kidney, liver, intestines, stomach, lymph nodes, spleen, thymus, genital organs, endocrines. In the kidneys, the order of intensity of pathological alterations was: proximal convoluted tubules, Henle’s loop, distal convoluted tubule, glomeruli, collecting tubules. Retrogressive changes of different degree and necrosis of the lining epithelial cells of tubules and endothelium and epithelium of glomeruli were the important lesions. Changes in glomeruli and Bowman’s capsule noticed in the higher dose group included shrinkage of glomeruli and presence of proteinaceous material in the capsular space. Eosinophilic granular casts and PAS positive bodies were present in the lumen of tubules. The necrobiotic renal changes were more intense when orchatoxin and aflatoxin were administered simultaneously. Hepatic lesions were mainly fatty infiltration, necrosis of hepatocytes and haemorrhage. The changes were most severe in combined toxicity. Mallory bodies and mild biliary hyperplasia were noticed in a few sections. Necrosis and subsequent depletion of lymphocytes wee the lesions in lymph nodes, spleen and thymus in some test animals. Degenerative changes were also noticed in testis, ovary, pituitary, adrenals and pancreas in experimental groups. In the combined toxicity group the pathological effect was more intense. At the ultra-structural level, the hepatcytes as well as the epithelial cells in the kidney showed severe changes. The cell organelles were either completely damaged or showed partial configurational alterations. Mitochondria showed changed in the density of matrix as well as disorientation and destruction of the limiting membranes and cristae. Cytolysosomes incorporating damaged cell organelles were abundant. Disaggregation of ribosomes and fragmentation of ER were noticed. In the glomerulus, there was destruction of the basement membrane and disruption of the regular arrangement of the foot processes of podocytes. In the cytoplasm of hepatocytes, Mallory bodies and lipid droplets were present. Varying degree of nuclear changes like clumping, condensation and disappearance of chromatin and fragmentation of nucleolus and nuclear membrane were observed. Changes occurred in the tight junctions of epithelial cells of bile ducts. Pathological alterations were more pronounced when ochratoxin was administered by the pwerenteral route. Oral administration of toxin also effected structural alterations which indicated that some fraction of ochratoxin escaped degradation in the rumen. From this study it became evident that aflatoxin potentiated the effect of ochratoxin. The structural damage to the cells might be due to the inhibition of oxidative enzymes which is reflected by the extensive ultra- structural alteration observed in the mitochondria and RER. Biochemical changes like high BUN and creatinine were evidently due to necrobiotic changes in the kidney. Interference in the synthesis of proteins due to damage of hepatic cells and escape of protein molecules due to alteration in the podocyte foot processes and basement membranes may account for the reduced serum protein levels. The nature of organellar destruction and configurational changes in the cells indicate the toxic potency of the mycotoxin on the biological system.

    Studies on purple top roll (PTR) of potato in Karnataka

    1. Survey for the incidence of potato purple top roll revealed that the disease was present in Bangalore, Belgaum, Dharwad, Hassan and Kolar districts of Karnataka. The average percentage of incidence varied from 8.94 to 50.07. The survey also indicated that there was low incidence of the disease in summer and high incidence in kharif and rabi. 2. The pathogen was transmissible by wedge, side and core grafting. 3. The disease was transmitted through dodder, Cuscuta chinensis Lam. 4. The disease was not transmissible through sap and aphids Myzus persicae Sulz. And Aphis gossypil Glov. 5. The etiological agent was transmitted by the leafhopper, Orosius albicinctus Distant and not by Hishimonus phycitis Distant. 6. The vector, O. albicinctus could acquire the pathogen in two hr and inoculate to healthy plants in 30 min. The percentage of active transmission was 35 – 40 and the minimum incubation period in the plant was between 20 and 25 days. Maximum percentage of transmission was obtained on 40th day after inoculation. 7. Characteristic symptoms of the disease was inward rolling of basal parts of top leaflets, purple/pink pigmentation, smalling of leaves, excessive proliferation of axillary buds, erect appearance, production of purple coloured, aerial tubers and stunting and dwarfing of plants. 8. The etiological agent infected tomato, tree tomato, Datura stramonium and D. metel by grafting. 9. The pattern of spread of the disease in the field was from external source in the beginning and later the spread was within. 10. The vector population was correlated with the disease incidence in the field. The increase in the incidence of the disease followed the increase in the vector population. 11. The leafhopper vector, O. albicinctus Distant was high in the months from late June to December and low from January to early June. 12. The high population of leafhoppers was correlated with low temperature and high humidity and low population was correlated with high temperature and low humidity. 13. The disease appeared 40 days after planting in the field and then it gradually increased reaching the peak by 90th day. 14. The pathogen was transmitted through tubers for two successive generations but to a lower percentage in the second generation. 15. Of the 36 cultivars/varieties of potato tested under field conditions, none were found to be resistant, However, the vars. JF 542, JF 4612, JG 676, JG 900, JC 750 and JE 812 had less than five per cent infection. Kufri Chandramukhi and Kufri Jyoti were highly susceptible having 48.3 and 47.6 per cent incidence, respectively. The highest average incidence of PTR in these cultivars in farmer’s fields was 35.85 and 26.03 per cent, respectively. 16. There was considerable loss in yield in many cvs. Viz., JC 750, BS/F 100, JC 182 and EM/F 2120. 17. The weight of tubers produced by the infected plants was always less than the tubers obtained from the adjacent healthy plants. The infected plants produced more number of smaller tubers which aggregated close to the main stem attached on short stolons. 18. Complete remission was obtained at 500, 1000 and 2000 ppm, incomplete remission at 100 and 250 ppm and no remission at 50 ppm of tetracycline given alternatively for 20 days.

    Influence of particle size of feed and plane of feeding on growth,physiological reactions and carcass characteristics of growing-finishing pigs

    Thirtysix Large White Yorkshire barrows averaging 70 days of age and 11.2 kg weight were assigned at random to six dietary treatments in order to study the influence of feed particle size and level of digestible energy on the performance, physiological reactions and carcass characteristics. They were individually housed and maintained on diets compounded mainly of dried sardines, dried tapioca, groundnut cake and rice bran till they were 8 months of age. The barrows were provided with the experimental diets having 100%, 90% or 85% of NRC standards of DE with feed particle size of less than one or two to three millimeter. It was observed that variations in feed particle size did not produce any significant effect on growth, physiological reactions and carcass characteristics of pigs under any of the treatments. At 8 months of age, the pigs under treatments 1 and 4 (100% DE of NRC; 2 and 5 (90% DE of NRC) and 3 and 6 (85% DE of NRC) had, (i) a final weight of 102.9; 99.4; 86.8; 92.3; 67.1 and 67.2; (ii) an average daily gain of 0.545; 0.524; 0.448; 0.473; 0.329 and 0.326 kg, and (iii) feed conversion ratio of 3.4; 3.5; 4.3; 4.2; 5.6 and 5.4, respectively. The pigs under treatments 1 and 4 reached 70 kg weight at an age of 197.5 days on an average, while pigs under treatments 2 and 5 reached the same weight by 208.5 days, and under 3 and 6 by 228 days. The digestibility of dry matter, organic matter, ether extract and crude carbohydrate in pigs under all the treatments was significantly (p <0.01) lower with decreasing levels of DE at 5 and 7 months of age. Although the digestibility of crude protein was significantly higher (p <0.01) in pigs that received 100% DE of NRC than 85% at 5 and 7 months of age, it was statistically of the same order with that of 90% at 5 months of age. Dietary treatments had no significant effect on the rectal temperature, heart rate, respiration rate and haemoglobin concentration of blood of the experimental pigs. The yield of dressed carcass was decreased from 72.3% to 68.8% or 64.7% with the decreasing body weight of pigs on 100%, 90% or 85% DE of NRC standards, respectively. The yield of ham was increased from 21.3% to 22.5% or 23.5% with the decreasing levels of DE, respectively. The length of carcass, eye muscle area and overall backfat thickness were significantly (p <0.01) lower as the level of DE was decreased. The fat percentage in dried eye muscle was almost uniform (22.3 to 24.9%) in pigs under different dietary treatments. The quality score for marbling, colour and firmness was significantly (p <0.01) in favour of diets that contained higher of DE. The feed cost per kg body weight gain of pigs under different dietary treatments or total cost of feed and labour at market weight of 70, 80 and 90 kg were determined. The cost of raising pigs from weaning to these weights on rations containing 90% DE of NRC standards and two to three millimeter feed particle size was found to be most economic. An equation W = 5.16 + LG2/11568 was evolved predicting live weight of barrows ranging from 8 to 144 kg, where W, is the weight in kg, L, is the length of body in cm, and G, is the girth in cm.
