Sweet)(UNIVERSITY OF AGRICULTURAL SCIENCES GKVK, BENGALURU, 2016-08-12) SURESH; Ramesh, SEarly elimination of un-productive crosses and handling of reasonably a large segregating populations derived from a few promising crosses will help enhance efficiency of crop breeding. Further, direct selection for economically important traits such as pod yield and its component traits has been less effective. The indirect selection for such difficult–to-breed traits based on easily observable/assayable/scorable traits is likely to be more effective. However, such a strategy requires a priori identification of traits strongly associated with difficult-to-breed traits. The combination of high quantitative traits means, absolute and standardized range, variances and frequency of transgressive segregants estimated in F2 and F3 generations was used as the criterion to assess and identify crosses with high breeding potential in terms of recovery of high frequency of pure-lines with desirable combinations of target traits in dolichos bean. The correlation coefficients of easily observable traits such as plant height, primary branches plant-1, racemes plant-1 and raceme length with dry pod and dry seed yield plant-1 were estimated to identify the traits which were significantly associated with later traits. Based on these criteria, the crosses, HA 10-8 × RIL 3-180 and FPB 21 × RIL 3-180 were predicted to have better breeding potential. The traits such as plant height, racemes plant-1 and raceme length were suggested for use as surrogates of dry pod and dry seed yield plant-1 for developing improved pure-line varieties of dolichos bean.ArticleItem Open Access DIAGNOSIS OF BOVINE TUBERCULOSIS IN WILD ANIMALS BY POLYMERASE CHAIN REACTION(2007-12) Vathsala, M; Swathika, MC; Jayathangaraj, MG; Gomathinayagam, S; Ramesh, S; Abdul Basith, S; TANUVASDiagnosis of Mycobacterium bovis is important from the public health perspective since control measures, treatment are very significant, particularly in wild animals. To diagnose M. bovis infection in wild animals polymerase chain reaction had been used, using the primer sequence of pnc A 8 and pncA 11 that are specific for M. bovis pnc A gene. A 744 bp PCR product was generated out of 3 samples that consisted of blood, caseated nodule, and lung tissues collected from hyena, bear and lion maintained at Arignar Anna Zoological Park, Chennai, Tamil Nadu and were identified as M. bovis. A positive control of M. bovis DNA procured from Tuberculosis Research Centre (TRC), Chennai, Tamil Nadu was used and a negative control of a healthy bovine blood was used. The present study demonstrates the use of PCR as a sensitive diagnostic gadget for diagnosing bovine tuberculosis (BTB) among wild animals.ArticleItem Open Access Enrofloxacin induced oxidative stress in kidney homogenates of broilers and its amelioration with antioxidants vitamin e and alpha lipoic acid(2018) Elamaran, A; Ramesh, S; Vairamuthu, S; TANUVASThe experiment was conducted to evaluate the effect of enrofloxacin induced oxidative stress in kidney homogenates of broiler chickens. Total fifty broiler chickens divided into five groups at the age of day 38 were used for this study. Oxidative stress was produced in the chickens with Enrofloxacin at an oral dose of 10 mg/kg b.w. In Enrofloxacin (10 mg/kg b.w) treated group, there was a significant elevation in the oxidative biomarkers like superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), reduced glutathione (GSH) and marked increase in lipid peroxidation (MDA) of kidney homogenates. Treatment with either alpha-lipoic acid (100 mg/kg b.w) or vitamin E (100 mg/kg b.w) significantly restored the antioxidant status in Kidney homogenates. Whereas, co-treatment of alpha-lipoic acid (100 mg/kg b.w) and vitamin E (100 mg/kg b.w) had significantly restored the antioxidant parameters which are comparable with the normal control birds. Thus the results of this study suggest that enrofloxacin induced oxidative stress in broilers at the therapeutic doses can be effectively ameliorated by treating with vitamin E and alpha-lipoic acid combination than treating with either of these drugs alone.Book chapterItem Open Access Ethno Veterinary Approaches for Treatment of Bovine Mastitis(Satish Serial Publishing House, 2015) Ramesh, S; Srinivasan, M.R.ThesisItem Open Access EXPLORING RECIPROCAL DIFFERENCES AND ASSESSMENT OF BREEDING POTENTIAL OF DOLICHOS BEAN (Lablab purpureus L.) CROSSES BASED ON TRANSGRESSIVE SEGREGATION AND USEFULNESS INDICES(University of Agricultural Sciences, Bangalore, 2022-12-08) SATISH HOSAKOTI; SATISH HOSAKOTI; Ramesh, S; Ramesh, SThe plant breeders routinely attempt a large number of crosses including reciprocal crosses to generate variability to implement selection to identify superior recombinant inbred lines (RILs) for use as pure-line cultivars. Assessment of reciprocal cross differences and breeding potential of crosses aids in the efficient use of limited resources by allowing early elimination of inferior crosses. This strategy increases the pace and efficiency of breeding dolichos bean, as is true in other crops. The combination of high quantitative traits (QTs) mean, absolute range (AR), standardized range (SR), absolute phenotypic variance (σ2p), phenotypic coefficient of variation (PCV), genotypic coefficient of variation (GCV), transgressive segregation index (TSI) and usefulness index (UI) estimated in F2 and F3 generations was used as the criterion to assess and identify crosses with high breeding potential in terms of recovery of high frequency of RILs with desirable combinations of target traits. Presence/absence of significant differences in QTs mean, AR, SR, σ2p, PCV, GCV, TSI and UI between pairs of reciprocal crosses were considered as evidence for the presence/absence of reciprocal differences. Based on these criteria, the reciprocal crosses of HA 4 × HA 5 and HA 4 × GL 201 were comparable for all four traits, suggesting that crosses could be affected in any direction in dolichos bean. The breeding potential varied with the trait and the statistics used. Hence, based on UI (a comprehensive quantitative measure), the cross HA 5 × HA 4 was identified more useful than the other crosses for grain weight plant-1.ArticleItem Open Access Expression of Biomarkers in Canine Mammary Tumours(Excellent Publishers, 2019) Muniappan, N; Ramesh, S; Hemalatha, S; Mohamed, Shafiuzama; Preetha, SP; TANUVASBiomarkers identifiable in tumours have potential use in early diagnosis, staging, prognosis and assessment of response to therapy. This paper describes the immunohistochemical expression of a few reliable biomarkers in post surgical canine mammary gland tumour (CMGT). Immunohistochemical expression of biomarkers in canine mammary tumours was carried out in forty three mammectomised tissue samples. The intensity and distribution of immunostaining of the biomarkers in histological types of CMGT was studied. PCNA showed weak nuclear staining in benign tumour and strong nuclear staining in malignant tumours. Bcl 2 showed weak cytoplasmic staining in benign tumours and moderate cytoplasmic staining in 10-20% of epithelial cells in malignant tumours. Cytokeratin 8/18 expression was strong in >50% of epithelial cells in benign and malignant tumours. Vimentin expression was specific with strong cytoplasmic staining in the stromal fibrous tissue, stromal fusiform cells, myoepithelial cells in tubules, spindle cells, chondroid tissue and lipocytes. CEA expression was negative in benign tumours. Malignant tumours expressed moderate membranous to cytoplasmic staining in 10-20% of luminal epithelial cells. MUC 1 expression was weak membranous staining in <10% of apical portion of luminal epithelial cells in benign tumours. Overexpression of MUC 1 was observed as moderate membranous and cytoplasmic staining in 10-20% of luminal epithelial cells in malignant tumours. HER-2 expression was weak membrane staining in <10% of luminal epithelial cells in benign tumours. Moderate membranous and cytoplasmic staining was observed in 10-20% of luminal epithelial cells in malignant tumours. COX 2 enzyme expression was negative in benign tumours. Moderate cytoplasmic staining in 10-20% of luminal epithelial cells, myoepithelial cells, stromal fusiform cells and stromal inflammatory cells was observed in malignancy. Hence the expression of biomarkers in tissues is of diagnostic and prognostic significance before initiation of treatment regimen in canine mammary tumours.ThesisItem Open Access FIRST- AND SECOND DEGREE STATISTICS - BASED GENETICS OF QUANTITATIVE TRAITS IN DOLICHOS BEAN (Lablab purpureus L. Sweet)(UNIVERSITY OF AGRICULTURAL SCIENCES GKVK, BENGALURU, 2016-04-10) SHIVAKUMAR, M. S; Ramesh, SFirst and second degree statistics-based genetics of quantitative traits, choice of the testers to discriminate advanced breeding lines (ABL) for their general combining ability (gca) and the relationship of parental genetic divergence (GD) as quantified by phenotype-based gca and cross legume species/genera transferable simple sequence repeat (SSR) marker alleles with hybrid heterosis and specific combining ability (sca) in dolichos bean were investigated at the Department of Genetics and Plant Breeding (GPB), University of Agricultural Sciences (UAS), Bengaluru during 2013-2015. While first-degree statistics suggested predominance of dominance and dominance × dominance-based gene interaction effects, second degree statistics revealed predominance of genes with additive effects in the inheritance of most of the traits. The estimates of additive genetic variance (σ2 A) and dominance genetic variance (σ2 D) and hence narrow-sense heritability were biased due to epistasis but the magnitude and direction of the bias varied with the traits. The ability of Low yielding testers (LYT) was better than that of high yielding testers (HYT) to discriminate ABL for their gca. While the, FPB 20 was desirable general combiner for dry pods plant-1, the variety HA 3 was a desirable general combiner for dry seed weight plant-1. One-hundred and twenty six (45.81 %) of the 275 cross legume species/genera SSR markers were transferable to dolichos bean and the extent of transferability varied with the cross legume species/genera. Among these 126, 83.33 per cent of Medicago truncatula, 78.57 per cent of green gram, 77.77 per cent of chickpea, 53.33 per cent of soybean and 33.33 per cent of cowpea SSR markers were transferable to dolichos bean. The conditional probability that a given cross with high heterotic/sca status is derived from parents of moderate GD was higher than that derived from parents of extreme GD. The study indicated the possible limits to parental GD for the occurrence of hybrid heterosis.ArticleItem Open Access GASTRIC IMPACTION IN OSTRICH CHICKS(2007) Hemalatha, S; Ramesh, S; Purushothaman, V; TANUVASImpaction is an accumulation of food or non-food items (Sticks, metal, glass etc.) in the proventriculus with the subsequent blockage of the opening into the gizzard causing a failure of food to move along the gastrointestinal tract , (Huchzermeyer, 1994, 1998). This paper reports cases of gastric impaction in ostrich chicks encountered at necropsy.ThesisItem Restricted GENOME BASED STUDIES ON PARVOVIRUSES AFFECTING DOGS AND CATS(2018) Mammen, Lancy; Parthiban, M; Shoba, K; Ramesh, S; Senthilkumar, TMA; TANUVASThe emergence of Canine Parvovirus and Feline panleukopenia virus in canine and feline population has prompted inputs particularly in the areas of diagnosis, as infection rates have been found to increase in the recent past globally and in India and early detection of CPV and FPV during such periods is very important to initiate appropriate diagnostic and prophylactic measures. Vaccines against CPV and FPV has at times resulted in vaccination failure due to introduction of newer antigenic variants.ArticleItem Open Access Gross, cytological, histopathological and Ultrastructural features of canine sweat gland adenocarcinoma: A case report(2021) Subapriya, S; Pazhanivel, N; Ramesh, S; TANUVASA 12 year old, male, Spitz breed dog was presented with a mass in the thigh region for tumour diagnosis. Samples were collected by Fine Needle Aspiration Biopsy (FNAB) for cytological examination. Tissue samples were collected post surgical excision for histopathological, immunohistochemical and ultrastructural investigation. Cytology of the mass revealed pleomorphic neoplastic cells containing round to oval nuclei and prominent multiple nucleoli. Histopathological examination revealed varying sized ducts and tubules containing pleomorphic neoplastic cells containing round to oval vesicular nuclei, prominent nucleoli and pale eosinophilic cytoplasm. Transmission Electron Microscopy (TEM) revealed tumour cells with features of anisokaryosis, multiple nuclei in a cell, margination of heterochromatin and prominent nucleoli with coarse chromatin. Based on the above findings the mass was diagnosed as sweat gland adenocarcinoma.ThesisItem Open Access IDENTIFICATION AND CHARACTRIZATION OF IMPORTANT PATHOGINS AND DETECTION OF PROBIOTIC BACTRIA FROM CAPTIVE REPTILES(TANUVAS, 2016) Dattatray Kisan, Ingole; Jayathangaraj, M.G.; TANUVAS; Gomathinayagam, S; Vijayarani, K; Ramesh, SA study was carried out in reptiles that were reared under captive conditions in captive wild animal places like Chennai Snake Park Trust, Guindy, Chennai; Arignar Anna Zoological Park, Vandalur, Chennai; Rajiv Gandhi Zoological Park and Wildlife Research Centre , Katraj, Punc; Nisargakavi Bahinabai Chaudhary Prani Sangrahalay, Sambhajinagar, Pimpri-Chinchwad, Pune and Mahatma Gandhi Prani Sangrahalay, Solapur.
