M.The present investigation was carried out at the Department of Biotechnology, ASPEE College of Horticulture and Forestry, Navsari Agricultural University, Navsari , Gujarat on "Micropropagation in ber cv. Gola" during the year 2008-2010. The surface sterilization of explants, treatment involving HgCI2 (0.1 %) for 3 minutes gave better sterilization of shoot tips and axillary bud from seedling and axillary bud from mature explants. Although, contamination was less in seedling explants, establishment of explants was higher in explants of mature plant. The use of mercuric chloride alone was more effective in reducing the contamination per cent as compared to sodium hypochloride. . . Maximum per cent establi shment of all type of explants viz. shoot tips and axillary bud from seedling explants and axillary bud from mature explants were observed on tre atm ent MS medium containing 0.4 mg!1 BAP. I MS medium supplemented with 2.0 mg/l BAP exhibited maximum multiplication rate of shoot with higher length of shoots in axillary bud from mature explants. In serial sub cultures, subsequently, multiple shoots and length of shoot (cm) were reported higher in the same treatment up to three sub cultures. Maximum proliferation of culture was recorded in 3% sucrose in the medium followed by 4% sucrose. Media pH had much influence on growth of regenerated shoots. Among the different pH levels tested, 5.S pH was found to be the best for maximum number of shoots and length of shoot followed by 5.7 pH. Maximum number of shoots, length of shoot and growth rate was observed at 3000 lux light intensity. In vitro rooting in regenerated shoots was found the best with full strength MS medium supplemented with 0.5 mg/l IBA, which recorded the maximum number of roots/shoot (6.S3) and length of root (6.73 cm). In vitro grown plant lets having (6-S em length of shoot) exhibited better survival of plantlets FYM + soil + sand (I: I: lv/v) potting mixture.
