The present study was conducted on liver of 12 pigs collected from local slaughter house, Ludhiana. The livers were thoroughly examined for any pathological lesions and rinsed with normal saline solution. Tissue samples from eight livers were fixed in 10% neutral buffered formalin, processed for paraffin block preparation by acetone-benzene schedule. The paraffin sections of 3-5 μm were cut with the help of rotary microtome and stained with different histomorphochemical stains. The cryostat sections of fresh frozen tissues of 10 μm thickness were cut at -23˚C and incubated in different substrates to localize phosphatases, oxidoreductases and Non specific esterases. It was revealed from present study thar left lateral lobe was significantly (p<0.05%) largest and quadrate lobe was shortest. Portal vein divided into three main branches; Right lateral vein, Right medial vein and left medial vein. The size of hepatocytes in periportal area (15.18 ± 0.28 μm) was significantly (p < 0.01) more than size of hepatocytes in centrilobular area (12.36 ± 0.25 μm). The number of mononuclear hepatocytes was significantly more (p < 0.01 ) in periportal area than centrilobular area whereas number of binucleated hepatocytes didn‟t vary significantly (p>0.01) between periportal and centrilobular areas. The average no. of Kupffer cells in periportal region (27.7 ± 0.76) was significantly more (p < 0.01) than centrilobular area (17.1 ± 0.72). The reaction for NMPS and glycogen was more in centrilobular hepatocytes, proteins were distributed homogeneously and lipids showed more reaction towards centrilobular hepatocytes. Periportal hepatocytes exhibited a strong activity of phosphatases and different oxidoreductases whereas non specific esterases activity was more in centrilobular hepatocytes.en_US
