dc.description.abstractThe research entitled “Enhancing efficacy of capsule based formulations and molecular characterization of local isolates of Lecanicillium lecanii (Zimm.) Zare and Gams” was carried out at Department of Entomology, College of Horticulture Bagalkot, UHS, Bagalkot during 2019-2020. The principle objectives of study were, 1. Enhancing the efficacy of capsule formulations of local isolates of L. lecanii. 2. Molecular characterization of local isolates of L. lecanii. The capsule formulations of L. lecanii was enhanced their efficacy against Myzus persicae by incorporating various additives. Chitosan added in the equal ratio to spore powder and talc in (1:1:1) resulted better efficacy in both laboratory and pot culture experiments, followed by spore powder and talc (1:1) and spore powder +talc + dextrin (1:1:1). Among the tested combinations, capsule containing spore powder + talc + chitosan (1:1:1) proved better efficacy against aphids when stored at refrigerated condition after 90 days of storage period followed by mud pot filled with sand and room temperature. The same formulation was tested after 90 days of storage period. The efficacy of formulations were tested against pot cultured cabbage seedlings against M. persicae under pot culture experiment and proved highest mortality of aphids when compared with commercial formulations of L. lecanii and NSKE at 5%. However chlorantraniliprole showed significantly highest mortality of aphids found to be superior over all the treatments. Molecular characterization of local isolates of L. lecanii was done using universal primers ITS-1 and ITS-4 as forward and backward primers, respectively. DNA sequencing resulted that, LL-1 groups with uncultured fungus group and LL-2 and LL-3 joined with other fungal endophyte group. Morphologically these isolates identified as L. lecanii by Agharkar research institute, Pune, whereas molecular study indicated that, these isolates belongs to other fungal groups. Hence, reconfirmation work to be carried out.
