Effect on hb strong in rumen profile in experimentally induced rumen dysfunction in buffaloes

Abstract
The high amount of polyunsaturated fatty acids in the plasma me mbrane of buffalo bull spermatozoa mainly attribute to its poor keeping quality. Vitamin L and C are two natural antioxidants with proven membrane protection effects. In the present study in vitro addition of these two compounds individually and in combination were evaluated. Semen samples were collected from six breeding buffalo bulls. Samples were diluted in Egg Yolk Citrate (EYC) and divided into four groups. Group I as control, Group II fortified with vitamin E@ I mg/ml, Group III fortified with vitamin Cq___-,01mM/m1 and Group IV fortified with combination of two in the same concentration as above. In group I motility varied from 66.19 ± 0.51 at zero hrs to 13.89 ± 0.63 at 72 hrs. The motility in group II, [II and IV were 18.99 ± 0.53, 15.72 ± 0.57 and16.74 ± 0.49 respectively at 72 hrs. Percent lire spermatozoa in group I ranged between 69.65 ±0.28 to 16.57 ± 0.49 from zero to 72 hrs. In group II, III and IV this value at 72 hrs were 23.59 ± 3.04, 17.97 ±0.45 and 20.36 ± 0.36 respectively. Lipid peroxidation was measured as malondialdehyde (MDA) produced in nmol/108 spermatozoa. In group I this value increased from 1.99 ± 0.26 to 8.7 ± 0.1 between zero to 72 hours. In group II, III and IV MDA level in nmol/108 spermatozoa at 48 hours were 7.47 ± 0.03, 7.93 ± 0.09 and 7.76 ± 0.05. At 72 hours these values were 8.14 ± 0.09 8.5 ± 0.11 and 8.45 ± 0.09. In group I spermatozoa superoxide dismutase activity ranged between 1.98 ± 0.09 at zero hours to 0.61 ± 0.02 U/108 spermatozoa at 72 hours. In group II, III and IV the activity of this enzyme was 0.83 ± 0.08, 0.65 ± 0.02 and 0.69 ± 0.07, respectively. Glutathione peroxidase in group I spermatozoa varied between 877.23 ± 21.7 U/108 spermatozoa to 175.26 ± 6.5 U/108 spermatozoa between 0 and 72 hours. At 72 hours the activity of this enzyme in group II, III and IV were 289.84 ± 38.6, 209.13 ± 9.8 and 224.45 ± 9.45 U/108 spermatozoa, respectively. The glucose 6-phosphate dehydrogenase in group I spermatozoa was 1.29 ± 0.05 U/108 at zero hours and 0.28 ± 0.02 U/108 spermatozoa at 72 hours. The-enzyme activity at 72 hours in group II, III and IV were 0.38 ± 0.02, 0.31 ± 0.02 and 0.33 ± 0.01 U/108 spermatozoa, respectively. These results suggested that vitamin E was a good membrane protector of buffalo spermatozoa in EYC at refrigeration temperature, whereas vitamin C alone or combination of the two was not as effective as single use of vitamin E.
