Abstract
Gatifloxacin is a newer, fourth-generation, 8-methoxy fluoroquinolone that exhibits broad spectrum of antibacterial activity than the older fluoroquinolones. The present study was designed to investigate pharmacokinetics of gatifloxacin following single dose intravenous and oral administration at the dose rate of 10 mg/kg of body weight, to determine effect of piperine coadministration at the rate of 15 mg/kg of body weight on the pharmacokinetics of the gatifloxacin at the rate of lOmg/kg of body weight and to evaluate safety and tissue distribution after repeated oral administration (10 mg/kg) of gatifloxacin at 12 hours interval for 14 days in broiler birds. Drug concentration in plasma was determined using High Performance Liquid Chromatography (HPLC). Pharmacokinetic parameters were calculated using noncompartmental approach. Following intravenous administration the therapeutically effective serum concentration of gatifloxacin > 0.36 fxg/ml was maintained for up to 12 hours. Based on the plasma gatifloxacin concentrations, various pharmacokinetic parameters like elimination half-life (t1/2β) (3.62 ± 0.03 hours), apparent volume of distribution (Vd(area)) (2.5 ± 0.025 1/kg), volume of distribution of drug at steady-state (Vd(ss)) (2.19 ± 0.015 1/kg), total body clearance (CIB) (0.48 ± 0.002 l/hr/kg), area under plasma drug concentration-time curve (AUC) (20.89 ± 0.101 µg-h/ml), area under first moment of curve (AUMC) (95.63 ± 0.78 µg-h2/ml) and mean residence time (MRT) (4.58 ± 0.027 hours) were determined.
