dc.description.abstractAn investigation was carried out on “Genetic variability and molecular characterization in China aster (Callistephus chinensis. [L.] Nees.)” during 2013-2014 at department of Floriculture and Landscape Architecture, Kittur Rani Channamma College of Horticulture, Arabhavi. Analysis of variance revealed highly significant difference among the 17 genotypes for all the characters studied. Narrow differences between GCV and PCV were observed for plant height and plant spread. Leaf area, plant spread, flower stalk length and number of flowers per plant were highly significant and positively correlated with flower yield per plant, suggesting the possibility of simultaneous selection for these characters. Path analysis for flower yield per plant revealed that number of branches, days to first bud initiation, individual flower weight, disk diameter of flower, number of flowers per plant, weight of ten flowers, number of leaves and flower diameter had high direct effects which indicated the possibility of increasing flower yield by selecting the genotypes for these characters directly. Molecular characterization was carried out to know the genetic diversity using PCR based SRAP and ISSR markers. Eleven SRAP primer combinations gave consistent banding patterns and were used to generate the SRAP fingerprinting. The primers combination Me5 + Em3, Me7 + Em4 and Me5 + Em2 were produced high polymorphism. Similarity matrix coefficient ranged from 42 to 66 per cent. Twelve ISSR primer combinations gave consistent banding pattern, among these UBC-810 and UBC-841 were produced maximum polymorphic bands. Similarity matrix coefficient ranged from 14 to 63 per cent suggesting lower to very high genetic variation within the China aster genotypes. Cluster analysis showed grouping of the genotypes based on source of collection (AAC-1 and AAC-2, Kamini, Shashank Poornima and Violet cushion), flower colour (AAC-1 and AAC-2, and Violet cushion and PG-Violet) and plant morphology (AAC-7, AAC-8 and AAC-9). From the study, it was found that genetic variation among selected China aster genotypes was high. The genetic distance information can be used for marker-assisted selection (MAS) and for the identification of diverse parents for future use in hybridization programme.en_US
