Hence, measures needed to alleviate fibrosis is crucial. The ameliorative effect of natural products, nanoparticles (especially liposomes) and antibacterial therapy in mastitis condition is evident from literature. Resveratrol, a polyphenol found in plants, has proven antioxidative, antibacterial and antifibrotic properties. Jamun (Syzygium cumini L.) is native and readily available in the country and its seed powder containing resveratrol has numerous biological health benefits. Cefquinome, a fourth-generation cephalosporin antibiotic is already approved for use in mastitis treatment. Therefore, the study was designed to assess the prophylactic effect of resveratrol, liposomal resveratrol formulation and Jamun seed powder in Staphylococcus aureus induced mastitic fibrosis goats in combination with cefquinome. In addition, the disposition kinetics of resveratrol and cefquinome in mastitic condition was also evaluated. Twenty-two lactating female Black Bengal goats (1.5-2 years age, 15-20 kg) were randomly divided into eight groups (Group 1 to 8) with three goats in each group (except Group 8 having one). Characterization of Liposomes containing resveratrol was done by Transmission Electron Microscopy (TEM), Dynamic Light Scattering (DLS) and Zeta potential estimation that revealed spherical shaped liposomes, around 200 nm size with zeta potential of -26.8 indicating good stability and lower chance of aggregation. Antioxidant parameters were found to increase and fibrosis biomarkers viz. Nuclear factor Kappa B p65 (NF-ΚB p65) and Transforming Growth Factor Beta 2 (TGF β2) decreased following jamun or resveratrol administration. The histopathological analysis too showed lesser degree of fibrosis in resveratrol or Jamun treated goats compared to positive control goats. The kinetic behaviour of resveratrol and cefquinome followed both one and two compartment open models in different groups and pharmacokinetic interaction was evident between the two drugs.
