    Clinico-therapeutic studies on bacterial mastitis in goats

    The lactating does in the University goat and sheep farm were screened for subclinical mastitis once in three months using the California mastitis test. The occurrence of subclinical mastitis was found to be 30.2 per cent. Statistical analysis showed no significant association between occurrence of subclinical mastitis and teat length whereas there was significant association between distance from teat tip to floor. Among 642 samples screened 194 samples were found to be positive by CMT. The arithmetic mean cell counts for each CMT score ranged between 0.736 Ā± 0.033 x106 and 20.417 Ā± 0.851 x106 cells/ml. Among CMT positive samples MWST and MAMP detected 62.89 per cent and 43.29 per cent as positive for subclinical mastitis. Comparison of screening tests revealed that significant positive correlation existed among the four tests namely CMT, MWST, MAMP and SCC. Comparison with culture results showed that score ā€˜3ā€™ of CMT score ā€˜3+ā€™ of MWST and grade 3 of MAMP reaction detected the maximum positive cases. CMT scores and SCC in bacteriologicaly positive samples showed significant association. Among the TIST positive milk samples 20 (44.44 per cent) were culture positive. Staphylococcus aureus was the most predominant isolate in both clinical and subclinical caprine mastitis. In vitro antibiotic sensitivity pattern revealed that chloramphenicol was the most sensitive antibiotic followed by ceftriaxone and ciprofloxacin. The isolated pathogens showed maximum resistance to sulpha. Comparison of treatment trials in 24 clinical goat mastitis cases using ceftriaxone and ciprofloxacin with 12 animals in each group revealed that clinical and bacteriological cure was better in the case of ciprofloxacin. Clinical and bacteriological cure was comparatively less in gangrenous mastitis cases. Eighteen Staphylococcus isolates from clinical mastitis cases and 23 Staphylococcus isolates from subclinical cases were typed by RAPD fingerprinting. Twelve different genotypes were obtained among which genotype c predominated in clinical mastitis whereas in subclinical cases b and i were the common Staphylococcal genotypes. Clinical and bacteriological cure rates were 100 per cent for RAPD type l in the ceftriaxone treated group and genotypes c and i in the ciprofloxacin treated group of animals. A possible relationship regarding the genetic make up of the different Staphylococcal isolates was elucidated from the phylogenetic tree generated from the RAPD fingerprints.

    Immunoprophylaxis against common dog tick using gut antigen

    The occurrence of tick infestation in dogs in the Corporation of Thrissur, Kerala, India, was studied in 1200 dogs during a period of one year from July 2004 to June 2005. Four hard tick species viz Rhipicephalus sanguineus, R. haemaphysaloides, Haemaphysalis bispinosa, H. bispinosa var. intermedia were identified. Significant difference in species wise prevalence was found on 124 tick positive dogs. The most prevalent species of tick infesting dogs was found to be R. sanguineus (8.58 per cent) followed by H. bispinosa (1.33 per cent), R. haemaphysaloides (0.33 per cent) and H. bispinosa var. intermedia (0.08 per cent). Three species of ticks namely R. haemaphysaloides, H. bispinosa, and H. bispinosa var. intermedia have been recorded in dogs for the first time from Kerala. The influence of month, season, age, gender and breed on the prevalence rate were observed. Prevalence rates were calculated by month, season, age, gender and breed. No significant difference (P>0.05) of tick infestation during different months was observed due to temperature, relative humidity, and rainfall. No clear pattern of seasonality was observed for R. sanguineus, which was present throughout the year. The different variables considered in the present study such as age and gender were not significantly associated with the presence of tick infestation in dogs. There was a highly significant (P<0.01) association between breed and the intensity of infestation with maximum intensity of infestation in German shepherd dogs. Sites of attachment of ticks were ear, neck, interdigital space, dorsum of the body, eyelids, perianal region, withers, thorax and hind limbs. Highly significant variation (P<0.01) was observed between attachment sites with maximum on ear (84.68 per cent) followed by neck and interdigital space. Tick-bite naive guinea pigs inoculated with gut extracts and Freundā€™s adjuvant revealed induced immunity against R. sanguineus expressed by altered feeding and fertility parameters consisting of the prolonged engorgement period, reduced engorged female weight and feeding efficiency index, prolonged pre oviposition period, reduced oviposition period, egg mass weight, egg rate conversion efficiency and larval mass. Except the incubation period all the reproductive and feeding parameters differed significantly compared to the control. Inradermal test done on immunised guinea pigs to assess cell mediated immunity revealed both immediate and delayed hypersensitivity reaction. Humoral immune response assessed by agar gel immunodiffusion (AGID) and passive haemagglutination (PHA) revealed that the former was less sensitive compared to the latter. Passive haemagglutination test detected anti tick antibodies as early as 14th day post immunisation. Peak titre 1:128 reached on the 35th day post immunisation.

    Clinical and ultrasonographic investigations of ascites in dogs

    Study entitled ā€œClinical and Ultrasonographic Investigation of Ascites in Dogsā€ was conducted in ten dogs. The study aimed at understanding the etiopathogenesis of ascites in dogs. The parameters observed were signalment, history and detailed clinical examination, electrocardiography, ultrasonography of liver, kidney and heart, course of illness, estimation of haemoglobin concentration packed cell volume(PCV), total plasma protein, albumin, A: G ratio, liver enzymes like alanine amino transferase (ALT) and alkaline phosphatase (ALP), protein content in ascitic fluid , ascitic fluid to plasma protein ratio, blood urea nitrogen (BUN), serum creatinine, sodium and potassium. Inappetance and lethargy were observed in dogs with liver diseases. Cardiac palpitation, loud heart sounds and strong femoral pulse were noticed in dogs with CHF. Non- specific and vague signs were noticed in dogs with nephrotic syndrome. Deep ā€˜Qā€™ waves in leads I, II and aVF, prolonged ā€˜QRSā€™ duration, S-T slurring, tall ā€˜Rā€™ waves, mild sinus arrhythmia and S1, S2 and S3 pattern were the abnormal ECG findings in dogs with CHF. No marked changes could be observed in the ECG of dogs with ascites of hepatic and renal origin. Ultrasonography of liver revealed hyperechogenicity of parenchyma, specks of hyperechogenicity and mildly echogenic gall bladder contents in three out of five dogs with ascites of hepatic origin. Two dogs had uneven and eroded borders along with hyperechoic liver parenchyma in dogs with ascites of hepatic origin. Nephrosonogram was unremarkable in all the ten dogs. Ultrasonographic findings and serum biochemical findings were coinciding with each other. Ultrasonography was an efficient tool in studying the changes of liver parenchyma and portal vasculature. ECG in cardiac diseases was complementary to echocardiography. Echocardiography was efficient in diagnosing DCM (two dogs) and HCM (one dog). All the dogs with liver diseases had mild to marked elevation in serum levels of ALT and/ or ALP. Hypoproteinemia and hypoalbuminemia were observed in dogs with liver and kidney diseases. Liver and kidney function tests were unremarkable in dogs with nephrotic syndrome and heart diseases. Treatment regimen involved administration of furosemide and/ or furosemide + spironolactone, silymarin, Liv- 52 Vet, enalapril, digoxin and prednisolone as the case may be. Six out of 10 dogs survived beyond 30 days following the therapy instituted. Nephrotic syndrome in dogs could be concluded by progressing hypoproteinemia especially hypoalbuminemia, low- protein ascites, negative ECG and echocardiographic findings and non- responsiveness to therapy. Nephrotic syndrome can be confirmed by biopsy and / or urine protein: creatinine ratio. Liver diseases can be confirmed and characterized only with biopsy.

    Comparison of different methods of pregnancy diagnosis in bitches

    With the object of comparing different methods of pregnancy diagnosis and to determine the early reliable method, a study was undertaken to investigate the efficacy of transabdominal palpation, ultrasound scanning and haematological profile during pregnancy at various stages of gestation in bitches. Sixty six apparently normal healthy bitches were included in the study. The bitches subjected to pregnancy diagnosis were grouped based on gestational period as 20 to 30, 31 to 40 and 41 to 65 days post breeding. The data obtained were compiled and tabulated.

    Medroxyprogesterone acetate as an aid to birth control programme in stray dogs

    The utility of Medroxyprogesterone acetate as an aid in the birth control programme of stray dogs was studied. Eighteen early proestrus bitches were randomly allotted to three equal groups (Group A Group B and Group C). Group A animals were administered with a single subcutaneous injection of 50 mg medroxyprogesterone acetate. Group B animals were administered medroxyprogesterone acetate orally at the rate of 10 mg per day for four days followed by 5 mg per day for another 12 days starting from the end of anoestrus. Six bitches of Group C acted as control animals. All animals in Group A and Group B responded to the treatment though one animal in Group A had returned to cycle in about 50 days. Reproductive cycle was controlled in all six animals of group A at a mean of 3.17 days and a mean of of 2.67 days in group B. Group A animals had a mean duration of control of 83.3 days while in group B it was 90 days. Vulval edema, vulval discharge, tail deviation reflex and interest in male decreased following treatment in Groups A and B while it seemed to be not changed or high in Group C animals. Detailed vaginal cytology was studied using Wright- Giemsa's stain. Cellular changes was characterized by a change from predominance of parabasal cells following treatment in Groups A and B while changes characteristic of oestrus cycle occurred in the control animals. There was significant difference in the mean percentage of parabasal cells between Groups A and B after treatment and during different stages of the oestrus cycle of the Group A up to end of dioestrus. Serum progesterone was estimated in all animals at fortnightly intervals. Animals in group A, B and C showed mean serum progesterone concentrations of 0.53 ng/ml, 0.57 ng/ml and 0.45 ng/ml during the beginning of proestrus. The mean progesterone in serum following treatment elevated in Group A to 27.83 ng/ml, while it maintained a low level of 0.55 ng/ml in group B and had a value of 4.43 ng/ml in group C which indicated oestrus phase. The progesterone level reached maximum concentration of 31.17 ng/ml on day 42 in group A and 49.00 ng/ml during dioestrus in group C. The values remained low in group B. Towards the end of experiment all animals had basal concentration of progesterone. It is concluded that reproductive control can be achieved effectively in bitches treated with medroxyprogesterone acetate by both oral and parenteral routes. Oral administration demand less technicalities and hence is more suitable for use in the field.

    Effect of dietary Cation-Anion balance on growth performance of broiler chicken

    An investigation over a period of six weeks was carried out to study the effect of dietary cation-anion balance on growth performance of broiler chicken. One hundred and sixty commercial day-old broiler chicks were randomly allotted to four dietary treatments viz., Tl, T2, T3 and 14 having four replicates each. The experimental diets were standard broiler ration with varying DCAB of 260 meq/kg (Tl, control), 210 meq/kg (T2), 310 meq/kg (T3) and 360 meq/kg (T4). Cation-anion balance was varied using sodium bicarbonate and ammonium chloride. Results indicated that the dietary treatments did not differ significantly in body weight, weight gain, feed conversion ratio, feed intake, water consumption and processing yields. Body weight at the end of six weeks was 2069, 2031, 2047 and 2020 g and the cumulative mean body weight gain was 2023, 1984, 2000 and 1973 g for Tl, T2, T3 and T4, respectively. Average daily feed intake was 159.39, 164.46, 166.65 atid 169.81 g and mean cumulative PGR were 2.04, 2.02, 2.04 and 2.09, respectively, for Tl, T2, T3 and T4. No significant difference was observed in serum minerals or biochemical parameters such as serum uric acid, cholesterol and glucose. Birds in T3 and T4 showed significantly higher (P<0.05) intake and balance of sodium than Tl and T2. Retention of chloride was significantly higher (P<0.05) for T3 and T4 whereas chloride intake was significantly lower (P<0.05) for T3 and T4. Overall evaluation of the results of the present study reveals that dietary cation-anion balance does not have a significant influence on growth performance of broiler chicken.

    Growth performance of broiler chicken fed on fermented fish waste silage ration

    An investigation spread over a period of six weeks was carried out to study the effect of replacing unsalted dried fish with fermented fish waste silage on nutrient utilization and growth performance in broiler chicken. One hundred and eighty day-old broiler chicks (Vencob) were divided into three identical groups having five replicates in each group with 12 birds in each replicate and allotted randomly into three dietary treatments viz., T1, T2 and T3. The three groups of birds were maintained on isonitrogenous and isocaloric rations in which protein of unsalted dried fish was replaced by protein from fermented fish waste silage at 0, 50 and 100 per cent levels. Birds were fed broiler starter rations up to four weeks of age and then they were switched to broiler finisher rations till the end of the experiment. The birds maintained on the three dietary treatments T1, T2 and T3 had almost similar growth rate (P>0.05) with a cumulative mean body weight gain of 1401.45, 1434.48 and 1491.31 g, respectively. The cumulative feed intake and cumulative feed conversion efficiency did not differ significantly (P>0.05) between treatments, the cumulative feed conversion efficiency being 2.12, 2.08 and 2.09, respectively for the birds of T1, T2 and T3. Nitrogen retention and balance were similar for the diets T1, T2 and T3. The retention and balance of calcium and phosphorus were also similar. Serum calcium, phosphorus, cholesterol, glucose, creatinine and uric acid contents were not significantly influenced by the inclusion of fermented fish waste silage. Serum triglyceride was significantly reduced (P<0.05) in T2 and T3 when compared to that of T1. Birds maintained on T3 registered significantly lower (P<0.05) ready-to-cook yield when compared to T1 fed on control diet. The cost of feed per kg gain of birds in the three dietary treatments was Rs. 20.15, 19.83 and 19.73, respectively. Overall evaluation of the results of the present study revealed that fermented fish waste silage could be used economically as a substitute for unsalted dried fish in the ration of broilers on protein basis, without any adverse effect on growth rate and feed conversion efficiency, while, the ready-to-cook yield tended to be lower in birds receiving diets with 100 per cent fermented fish silage.

    Post-hatch development of preen gland in the duck (anas platyrhynchos)

    The study on the post-hatch development of the preen gland in ducks was conducted using 44 ducks from the day of hatch to 150 days of age. The project was taken up to trace the structure and development of the glands and their relationship with the age and body weight. After recording gross relations and measurements, the material was fixed using various fixatives for studying the cellular details, arrangement of cells, connective tissue framework, micrometry and histochemistry. The preen gland was a paired organ with two gland and a common cylindrical papilla together formed a ā€˜Vā€™ shaped structure. Each gland was pear shaped and pale yellow in colour, in fresh state. They were located on the dorsal surface of the pygostyle. The two glands had separate ducts. The uropygial circlet was seen at the tip of the papilla. The glands were vascularized through a pair of branches from the caudal artery and innnervated through the medial caudal nerve. The weight of the preen glands increased progressively from the day of hatch to 150 days of age. This weight was positively correlated with the age and body weight. The proportion of the gland weight to body weight showed a decreasing trend. The right gland was slightly heavier, longer, wider and thicker than the left. The length, breadth and thickness were positively correlated with age and body weight. Simple, branched, tubular and holocrine type of glands was covered by highly vascular connective tissue capsule composed of collagen and reticular fibres. Elastic and smooth muscle cells were absent. The secretory tubules showed two zones, an outer zone or zone I, near the capsule and an inner zone or zone II, towards the primary cavity. The epithelium of the tubules consisted of basal, intermediate and transitional cell layers. The papilla had two ducts, which were lined by glandular epithelium initially and keratinized type of stratified stratified squamous epithelium at the tip. The glandular epithelium was surrounded by longitudinally arranged smooth muscle fibres and skin. Lamellar corpuscles and circlet feather follicles were noticed in the papilla. Capsule, trabeculae and the parenchyma were PAS positive. Acid mucopolysaccharides and glycogen were not detected in the gland. Lipids were evident uniformly in all the cell layers. The acid phosphatase activity was moderate in the basal and intermediate layers and strong in the transitional layer. The alkaline phosphatase activity was moderate in the basal and intermediate layers and weak in transitional layer of outer zone. It was moderate in the basal and intermediate layers and intense in the transitional layer of inner zone. Micrometrical findings on the capsule thickness, width of outer and inner zones and the primary cavity increased with the advancement of age.
