    Genetic variability, path analysis and stability parameters in sesame

    Biometric analysis in a varietal collection of sesame was undertaken to study the genetic variability, correlations, path analysis and stability parameters. One hundred sesame types were evaluated in replicated trials at Vellayani in uplands during rabi and at Kayamkulam in rice fallows during summer. Genetic variability and correlations were estimated and path analysis worked out independently as both the locations. Location trials for estimating stability parameters were conducted at three places viz. in uplands during rabi at Pattambi and Vellayani and in rice fallows during summer at Kayamkulam. Large values for genotypic coefficients of variation were obtained for characters such as number of capsules on branches, number of capsules perplant, number of capsules on main stem and number of branches during rabi as well as summer. The lowest genotypic coefficient of variation was obtained for number of days to maturity during both rabi and summer. High values of heritability were recorded by seed protein content , seed oil content, height upto first capsule and weight of 1000 seeds under both conditions.

    Influence of applied nutrients and stage of harvest on the yield and physicochemical properties of essential oil of palmarosa (Cymbopogon martini Stapf var. motia)

    Palmarosa, an essential oil crop introduced in Kerala, from Maharashtra, nearly two decades ago, is spreading steadily la the pialas and midland regions of North Kerala. Several agro-techniques have yet to he standardised for the commercial cultivation of this crop in the State. The present studies were undertaken at the Aromatic pnfl Medicinal Plante Research Station, Odakkaly during 1980-'8 4 to obtain information on the nutrition of palmarosa, optimum harvest intervals to ensure maximum herbage and oil yields and the factors influencing the quality of oil. The treatments in the major field experiment (1980-'82) consisted of three levels of N, and KgO, each at 25, 50 And 75 kg/ha along with six intervals of harvest at 40, 45, 50, 55, 60 and 65 days. The total number of treatment combinations wore 162 in a 3^ x 6 confounded asymmetrical factorial design. Tho main experiment was continued for another two yoaro limiting tho observations, to the yield of herbage and oil. This was then followed by an observation trial with intervals of harvest longer than tho maximum of 65 doye tried in the main experiment. In the main experiment the herbage yield vns significantly increased by application of P2°5 ^2°* Nitrogen did not have any eignifioant offeot on herbage yield poaeibly due to tha medium level etatua of soil N in the ABSTRACT ^erimental plots. The oil yield was Bignifioantly enhanced by P205 application at 50 kg/ha. Different levels and K showed no significant influence on the yield of oil. Harvest intervals showed significant influence on herbage yield, oil yield and oil content and the maximum value was recorded by the 65 day interval. A path analysis of the herbage yield with related cnaracters has shown that the height of the plant is the most important yield attribute influencing directly the yield of herbage. Path analysis of oil yield showed that oil yield is mainly dependent upon herbage yield. The direct effects on oil yield by yield attributes are found to be in the decreasing order of number of tillers with inflorescence, height of the plants and length of inflorescenoe. Maximum indirect affect via herbage yield is expressed by the height of the plant and length of inflorescence. All these directly and indirectly contributing factors arc soon to be markedly influenced by tho applioation of phosphorus and intervals of harvoat* Herbage yield and oil yield for various harvest lntorvalo obtained In the flrot two years wore fit tod In a Cobb-Dcughlua reoponoo function and tha expooted values «leulat«d were found to be very olooe to tho actual observed values• In thethird and fourth year of the experiment, the ghest herbage and oil yields were recorded by 60 day interval followed by the interval of 65 days, A quadratic Sanction for the data pooled over for four years was fitted to see if the data show a diminishing return with an increase in harvest interval and it was found that the optimum herbage yield and oil yield were obtained when the harvest was done at 62 and 63 days interval respectively. The final observational trial also indicated that the herbage and oil yields were maximum for a harvest interval of 65 days beyond which it decreases. Increase in the levels of both N and P tended to increase the content of geraniol and to decrease the content of geranyl acetate which are the price determining quality attributes of the essential oil. But the maximum interval of harvest viz., 65 days tried in the main experiment was not sufficient for moot of the physico-chemical properties of oil ouoh os specific gravity, refractive index, geraniol content, geranyl acotato content etc. to reach the minimum limit prescribed by 131. The observational trial showed that the oil obtained at intervale of 95 days and above satisfied the 131 specification with respect to all the phyalco-ohemioal properties of the oil. However, at thle herveat Interval the yield Itself wae considerably depressed compared to the yield at 65 day Interval. Th, eoonomloe worked out for different Intervale of harveab baaed on the ourrant market prloe of the oil paeein« as above the I SI limits as fe. 240/- per kg (oil from harvest intervals at and above 95 days) and those below them at Fs.220/- per kg (oil from harvest intervals of 55, 65, 75 and 85 days) it has been found that a wider cost benefit ratio and net return per rupee investment are obtainable for harvest intervals ranging from 65 to 85 days. However, 65 day interval has the advantage of early returns from the investment. Application of 50 kg PgO^/ha in view of its significant effect on herbage yield, oil yield and oil quality increase the net profit per rupee invested for the harvest interval of 65 days. The average removal o f II, P, K, Ca and Mg from the s o i l by palmarosa per h ecta re per year lias a lso been worked o u t .     Postnatal development of testis and epididymis, semen characteristics and fertility of brown- swiss crossbred bulls

    A systematic investigation involving the use of 24 Brown Swiss crossbred bull calves and 7 adult bulls with 62.5% exotic inheritance varying in age from 0- 360 days and 22-40 months respectively, resred and maintained at Indo-Swiss Project, Mattupatty, Kerala, was carried out in order to (a) trace the postnatal growth and development of the testis and epididymis and (b) assess the semen characteristics such as volume, initial motility, sperm concentration, live sperm count and incidence of abnormal sperms; As a corrolary to and as an integral part of this comprehensive study, 6679 semen collection particulars from the records maintained at the Indo-Swiss Project, Mattupatty and 7586 insemination data at the Crossbreeding Research Centre, Muvathuphza werecritically scrutinised to deduce freezability/discard rate of the ejaculates and the fertility potenoy of the bulls, respectively. The materials used, the salient observations made and the valid inferences drawn aare given below: Twenty four pairs of testis and epididymis collected from the bull calves were processed to study the postnatal growth and development. The pattern of growth of testis and of the epididymis was found to be curvilinear. The testicularand epididymal weights were observed to be significantly (p<0.01) correlated with the age, and body weight of the animals. A highly significant (p<0.01) correlation between epididymal weight and testicular weight was also observed. The growth rate of the seminiferous tubules appeared to be of sigmoid type. Formation of lumen in the seminiferous cords was initiated at 150 days and completed, at 180 days of age. The transformation of gonocytes to spermatogonia occurred at 150 days of age. The formation of primary spermatocytes, spermatid and spermatozoa cocurred at the age of 150 days, 300 days and 300-360 days, respectively. The process of transformation of supporting cells was initiated at 120 days and completed at 180 days of age. The transformation of immature Leydig cells to mature cells was initiated at 150 days and completed, by 180 days of age. There were distinct regional differences between the caput, the corpus and the cauda epididymis in respect of the diameter of the tubules, height of the epithelium and the degree of transformation of Epithelial lining from simple columnar to pseudostratified type. The distal part of the ductus epididymis seemed to have attained the adult size at an earlier age than the proximal. Sperms were seen in all the three regions at about 300 days. The mean values of ejaculate volume, initial motility, sperm concentration, live sperm count, sperm head abnormalities, tail abnormalities and proximal protoplasmic droplets were found to be 2.65 ± 0.84 ml, 66.24 ± 0.87%, 1599.64 ± 55.371 milions per ml of semen, 72.55 ± 1.310%, 4.91 ± 0.439%, 0.65 ± 0.045%, 6.47 ± 0.775% and 1.20 ± 0.143%, respectively. Significant (p<0.01) variations between bulls were observed in respect of sperm head abnormalities, tail abnormalities and proximal protoplasmic droplets and between months, in respect of ejaculate volume, sperm head abnormalities, free loose head, tail abnormalities and proximal protoplasmic droplets. Seasonal variation (p<0.05), however, was evident only in the incidence of tail abnormalities. Variations due to age of bulls were found to be significant (p<0.01) in the casse of ejaculate volume, sperm concentration, free loose head, middle piece abnormalities and tail abnormalities. The freezability of sperms expressed in terms of per cent of post-thawing motility was found to be 36.86. There was no seasonal effect on the freezability of sperms. The freezability rate increased significantly (p<0.01) with advancement of age of bulls. The overall rejection rate of the ejaculates was noted to be 39.80%. Most of the rejection (71.75%) was done before freezing. The pre-freezing rejection was found to be 28.55% of the total ejaculate. The post-freezing discard to be 15.74% of the frozen samples. The rejection rates of the ejaculates differed significantly (p<0.01) between months/seasons/age of the bulls. The rate of rejection was found to decrease with advancing age. The overall fertility rate of the bulls was found to be 56.06%. There were significant (p<0.01) differences in the fertility rate between months/seasons of insemination and between age of the bulls. As between bulls also, there was a significant variation (p<0.01) in the fertility rate. The significance of the observations inferences indicated above and their relevance to the implementation of cross breeding programme in Kerala were discussed briefly.

    Pathology of the reproductive organs in experimental hypothyroidism in goats

    A survey study was conducted to assess the role of hypothyroidism in the etiology of reproductive disorders in goats. Lowered functional activity of the thyroid was associated with postpartum anoestrum, repeat breeding and delayed puberty. An experimental model of controlled hypothyroid state was induced in goats, using different dose regimes of thiourea with the objective of studying the sequence of physiopathological changes in hypothyroidism and its influence on reproduction. Clinically healthy young and adult cross-bred goats of both sexes were employed for the study. The animals were divided into control, Treatment I (T1) and Treatment II (T2) groups. Groups T1 and T2 were administered thiourea orally at the rate of 50 mg/kg and 100 mg/kg body weight respectively. Clinical symptoms and growth rate were studied. Protein bound iodine, total serum protein and serum cholesterol were estimated. Haematological data were collected and the semen characteristics were evaluated. On the 90th day of the experiment both control and experimental animals were sacrificed leaving at least one animal in each group. These animals were maintained without treatment with thiourea for a further period of 90 days and sacrificed. Cross lesions and histopathological changes in tissues were recorded. Weakness, lethargy, depression, reduced feed intake, subcutaneous oedema of varying degree, loss of libido in males and anoestrum in females were the important clinical features observed. There was stunting of growth and apparent reduction in weight of the animals dosed with thiourea. On discontinuation of treatment with thiourea all the clinical signs manifested disappeared and the animals gained weight progressively. There was significantly decrease in the protein bound iodine, and increase in the total serum protein and cholesterol level in all the animals dosed with thiourea. The values reached the normal level on discontinuation of treatment with thiourea. Microcytic hypochronic anaemia was evident in hypothyroid goats. There was improvement in haemogram values on withdrawal of thiourea administration. Reduction in quantity and quality of semen was observed in hypothyroid goats. The semen appeared normal in quality and quantity on discontinuation of thiourea administration. Gelatinisation of subcutaneous fat and hypertrophy and dilation of the left verntricle were the common findings at autopsy. There was significant increase in the relative weight of the thyroid, pituitary and the adrenal gland. Histologically the thyroid glands exhibited varying degree of hyperplasia and hypertrophy. Predominant histological changes in the pituitary was hyperplasia and hypertrophy of basophil cells and degenerative changes in the acidophils. Adrenal gland showed hypertrophy and depletion of fat in the zona fasciculata. On discontinuation of treatment with thiourea all the lesions disappeared. A significant decrease in the relative weight of the testis and epididymis was observed in all the goats dosed with thiourea. Histologically the seminiferous tubules were small and lined by single layer of spermatogonial cells. Some tubules contained degenerated and desquamated cells without any evidence of sperms. The tubules of the epididymis were small in size and the lining cells showed deciliation and desquamation. These degenerative changes were found to be reversible on discontinuation of treatment with thiourea. The accessory sexual glands, the seminal vesioles and the prostate weighed less in hypothyroid goats. Histologically the cells lining the glands were small inactive, vaculated and hyalinised. After discontinuation of administration of thiourea the reparative changes were almost normal. There was significant decrease in the relative weight of the ovary and uterus in all the animals dosed with thiourea. Histologically ovaries were found to be inactive and only few scattered immature follicles were seen. Uterine glands were few and non secretary type. The lining epithelial cells did not show active mitosis. On discontinuation of treatment with thiourea the weight of the ovary and uterus increased and histologically ovaries and uterus were found to be normal. Kids and female goats were more susceptible to the effects of hypothyroidism. The significant pathological changes observed in the reproductive organs have clarified the importance of hypothyroidism in inducing sub-fertility and infertility in goats. The reversible nature of the lesion was also proved.
