Studies on Genetic Diversity in natural Watermelon (Citrullus lanatus Thunb.) Populations of Rajasthan
(DEPARTMENT OF BIOTECHNOLOGY COLLEGE OF AGRICULTURE RAJASTHAN AGRICULTURAL UNIVERSITY BIKANER (RAJASTHAN), 2009) Jitendra Kumar Bhakhar; R. A. Sharma
The present investigation was carried out to estimate genetic diversity at molecular level (RAPD) for twenty three genotypes of watermelon (Citrullus lanatus Thunb.). RAPD analysis was conducted using 14 random primers of which eight primers produced 41 bands, 39 being polymorphic, generating 95% polymorphism. Jaccard similarity index based on RAPD revealed 49.6% average similarity with a range of 28% to 86%.

Development of New Mapping Population and Marker- Assisted Improvement of Iron and Zinc Grain Density in Pearl Millet [Pennisetum glaucum (L.) R. Br.]
(Department of Biotechnology, COA, SKRAU, Bikaner) Sushil Kumar; Dr. Govind Singh
Pearl millet [Pennisetum glaucum (L.) R. Br.] is a multi-purpose grain/fodder crop of semi-arid regions where many of the world’s undernourished people live. Pearl millet grain can be biofortified by selective breeding to help alleviate micronutrient deficiencies among its grain consumers, especially in developing countries where Zn and Fe micronutrient deficiencies are common.

Construction and analysis of SSH library of early heat stress induced genes of Vigna aconitifolia (Jacq.) Marechal
(2012) Sakshi Rampuria; Dr. R. Sharma
Amongst 100 genotypes accessed for their heat tolerance level 9% were tolerant accessions, 28% were moderately tolerant accessions, 46% were moderately susceptible and 17% were susceptible. RMO-40 showed high degree of tolerance towards heat stress and was used for SSH cDNA library construction. Heat induction was carried out by exposing 14 d old seedlings to elevated temperature of 42oC for 5 min.

Study of comparative efficiency of Agrobacterium mediated and direct transformation through biolistic gun in chilli (Capsicum annum L.) cv. Mathania
(Department of Biotechnology COLLEGE OF AGRICULTURE SWAMI KESHWANAND RAJASTHAN AGRICULTURAL UNIVERSITY, BIKANER, 2010) MOHAMMAD RIZWAN; R.Sharma
A protocol for regeneration and genetic transformation has been established for chilli (Capsicum annuum L. cv. Mathania). High frequency regeneration of shoot buds from cotyledonary leaves and hypocotyls was achieved with Murashige and Skoog ’s (MS) medium supplemented with 10 mgL-1 BAP and 1 mgL-1 IAA. Elongation of shoots bud and subsequent rooting was obtained on MS medium supplemented with 1 mgL-1 IAA. A disarmed strain of Agrobacterium tumefaciens EHA 105 carrying a binary vector plasmid p35SGUSINT has been used for transformation.

Molecular Investigation on Genotypic Variations for Drought Tolerance in Pearl Millet
(DEPARTMENT OF BIOTECHNOLOGY COLLEGE OF AGRICULTURE, BIKANER SWAMI KESHWANAND RAJASTHAN AGRICULTURAL UNIVERSITY BIKANER (RAJ.), 2009) Riva Mathur; N.K. Gupta
The present investigation was undertaken to find out the effect of PEG induced water stress on morpho-physiological characters, antioxidants and gene expression of 32 genotypes of pearl millet. The PEG-6000 (15%) treatment was given to 7 days old aseptically grown pearl millet seedlings in test tubes containing ¼ M.S media, under laboratory conditions. Observations were taken after three days of PEG treatment.

Identification of Heat Related ESTs in Moth bean through Suppression Subtraction Hybridization
(Department of Biotechnology COLLEGE OF AGRICULTURE, BIKANER SWAMI KESHWANAND RAJASTHAN AGRICULTURAL UNIVERSITY BIKANER-334006 (RAJ.), 2011) KARUN GURJAR; Ramavtar Sharma
Moth bean (Vigna aconitifolia) represents an extraordinary source of genes that control traits of agronomic importance such as drought and heat tolerance. Seven day old seedlings grown at 27° C could tolerate a rise in temperature up to 42° C however most genotypes dried at 47° C after 6 h of treatment. Genotypes showed differential response to tolerance for 47° C for shorter periods (3h). Based on screening for heat tolerance variety RMO-40 was considered for molecular study of heat.

Effect of Post Anthesis High Temperature Stress on Molecular Mechanisms in Contrasting Wheat Genotypes
(DEPARTMENT OF BIOTECHNOLOGY COLLEGE OF AGRICULTURE, BIKANER SWAMI KESHWANAND RAJASTHAN AGRICULTURAL UNIVERSITY BIKANER (RAJ.), 2011) OMPRAKASH CHHAPOLA; N.K.Gupta
The present investigation was undertaken to find out the effect of high temperature stress on morpho-physiological characters, antioxidants and gene expression of four genotypes of wheat. The seeds of these genotypes were grown on Nov.15, 2010 (for normal sowing) and Dec. 15, 2010 (for late sowing) in the field. These genotypes were sown by employing standard package and practices recommended for this region. Results showed that there were significant variations in photosynthesis, transpiration, leaf conductance, leaf temperature and relative water content on account of late sowing.
