ThesisItem Open Access CHARACTERISATION OF RICE VARIETIES THROUGH MORPHOLOGICAL CHARACTERS,CHEMICAL TESTS AND ELECTROPHORETIC TECHNIQUES(Professor Jayashankar Telangana State Agricultural University, 1999-11) VIJAYA LAKSHMI, B.; RAGAIAH, B.Varietal identification is essential forseed certification inthe seedindustry. To realise the expected dividends from the released varieties, maintenance of genetic purity is utmost importance in seed productionprogrammes. The current methodology utilised in seed certification isveri fication in the grow out tests which is time consuming. Hence, thereisaneedtodevelopquickandreliablelaboratoryteststodeterminevarietal purity. Mostofthemorphological characters ofthevarieties arecommonhence a simple criteriacannotbe usedforvarietalidentification. Varieties canbeidentifiedbytakingsomeofthemorphologicalcharactersandwhere everthe characters are common we should take the help of chemical tests and or electrophoratic Poly'movPhtsm In the present study, an attempt was made to characterise and identify different rice genotypes which are in demand and active seed productionchainbasedonmorphologicalcharacters(leafcolour, anthocyanin colouration of leaf tip and margins, penultimate leaf blade pubescence, flag leaf curvature, . colour of stigma and kernel shape). Reaction ofseeds and/or seedlings to different chemicals (phenol, modified phenol, NaOH, KGH, FeSO4, peroxidase, GA3 and 2, 4-D test) andelectrophoretic banding patterns ofseedproteins. Seedkeys havebeendevelopedbasedonmorphologicalcharacters and response of seeds and/or seedlings to chemicals to veiify genetic purity ofseeds onroutinebasis in seedtesting laboratories. g Theelectrophoreticbandingpatternofseedproteinsforeachofthe genotype was unique and proved to beareliable and quick method for identification.
