One Trichoderma spp. was isolated from each agroclimatic zones usingPotato dextrose agar medium and purified. These isolates werecharacterized as T harzianum strains. Pure culture of the ten Trichoderma harzianum representing each zone along with a reference culture was maintained in the Department of Biotechnology, were separately grown in Potato dextrose broth medium. Molecular diversity of these isolates was characterized by using SDS-PAGE and RAPD markers. SDS-PAGE analysis revealed thatRmvalue of the bands ranged from 0.0246 and 0.995. In RAPD a total of 125 bands were scored out of which 100 bands were found to be polymorphic (80.00%). Statistical analysis of SDS-PAGE andRAPD data enabled the classification ef lOT harzianum isolates in to two major groups. The protein profile data and RAPD banding pattern of these isolates closely separate isolates from differentzones. Apot culture experiment was carried out to study germination, growth, biochemical and histochemical parameters on the medicinal plant Nigella sativa under glass house conditions. Nigella sativa plants inoculated with the T harzianum strains recorded increased germination, plant height, number of branches and biomass compared to uninoculated plants. Biochemical parameters include chlorophyll content, nitrogen, phosphorous, total sugars, reducing sugars and total soluble proteins. Among the 10 strains inoculated, the T harzianum strain isolated fromthe zone6 (SDT) recorded the highest plant growth, biochemical and histochemical parameters, which is followed by the strains isolated from other zones. The least biomass, biochemical and histochemical parameters were recorded in uninoculated plants.
