ThesisItem Open Access IN VITRO HYBRID EMBRYO RESCUE FROM INTERGENERIC CROSSES IN PAPAYA(2021-01-21) NAVEEN KUMAR, V M.; Dr. NANDEESHA, P.Papaya, Carica papaya L., is one of the major fruit crops cultivated and consumedin tropical and sub-tropical areas. The most destructive disease in C. papaya is papaya ring spot, caused by Papaya Ringspot Virus (PRSV-P). Interspecific and intergeneric hybrids production is suitable for transfer of desirable genes from wild species into cultivated species. Hybrid embryo rescue is the useful biotechnological method to produce hybrids which have pre or postfertilization incompatibility. Crosses were made between Arka Prabhath and PRSV-P resistant Vasconcellea species cauliflora and cundinamarcensis to produce intergeneric hybrid. Different matured fruits such as 30,60,90,120 and 150 days are collected and immature zygotic embryos were cultured in different medium such as MS, MS+ B5, WP and ER with growth hormones IAA and BAP at variable concentration. Days taken for embryo initiation and proliferation were recorded accordingly. Embryo viability test using 2,3,5- triphenyl tetrazolium chloride in different concentration of 0.1 ,0.5, 1.0 % were used in intergeneric seeds. The seeds of intergeneric hybrids were kept for incubation in germination chamber with the controlled cycling temperature of 22º C for 16 hours followed by 32º C for 8 hours to get better germination. After the seeds starts germinated, the germination percentage was recorded as 43.75 %. The seeds were transferred to poly bags for hardening. Genomic DNA were isolated for the hybridity confirmation of intergeneric cross made from parents Arka Prabhath and wild Vasconcellea species using SSR markers. The banding patterns of the parents and progenies were compared to test the hybridity.
