SinghThe present investigation entitled “In-vitro propagation of lasora (Cordia myxa Roxb.) using nodal segments” was conducted in tissue Culture Laboratory of the Department of Horticulture, Institute of Agricultural Sciences, Banaras Hindu University, Varanasi during 2014-2016 with a view to study the effect of different growth regulators on nodal segment proliferation and root regeneration to develop protocol for micro-propagation of lasora. A protocol for micro-propagation of lasora from nodal and inter nodal explants has been developed. Direct organogenesis method was found to be better than indirect organogenesis method in terms of per cent success of cultures. Nodal segments were found to be better than shoot tip and inter nodal explants. Nodal segments were surface sterilized and cultured on MS medium supplemented with different concentrations of growth regulators for shoot proliferation and root regeneration. According to the results obtained in this study, best surface sterilants for lasora explants includes HgCl2 (0.1%) for 10 minutes. [BAP (3.5 mg/L) and NAA (0.07 mg/L)] was found to be best for shoot proliferation encompassing all aspects, i.e. sprouting percentage, number of shoots, shoot length and leaf number. Among IBA and NAA, IBA alone with the concentration of 2.0 mg/L was noted the best treatment for root induction spreading over the aspects of per cent root regeneration, days to root initiation, root number and root length.
