Abstract
A total of 135 samples (50 liver, 50 tracheal and 35 faecal) were subjected to AGPT, CIEP and ELISA for direct detection of avian adenoviruses (AAV) in tissue suspensions. In AGPT, 9 liver and 1 tracheal samples, where as in CIEP, 11 liver, 11 tracheal samples and 4 faecal samples showed precipitin line with CELO-virus chicken antiserum. The plate-ELISA using double antibody sandwich method revealed 21 liver, 13 tracheal and 4 faecal samples having positive absorbance value. Thirty eight isolates comprising of 17 isolates from 50 different outbreaks of IBH, 16 isolates from 50 different cases of respiratory infections and 5 isolates from 35 cases of egg-drop syndrome were obtained in chicken embryo liver (CEL) cell culture. The primary identification of isolates were made on basis of characteristic cytopathic effects (CPE) including rounding, aggregation, cytoplasmic vacuolation, and degener¬ation of cells by 72 hours P.I. The presence of basophilic intranucelar inclusion bodies was also demonstrated. Further confirmation of isolates was made on the basis of their resistance to chloroform and serological identification using AGPT,CIEP and ELISA-test.It was found that ELISA was the most sensitive test which could detect all the 38 isolates,followed by CIEP 31 isolates and AGPT14 isolates
