NATH, AMARJIT K.TAANK, RICHA2017-02-172017-02-172010http://krishikosh.egranth.ac.in/handle/1/5810001405ABSTRACT Eighteen different grain legume cultivars of Himalayan region were analyzed for -amylase inhibitor activity. -Amylase unit inhibited per gram seed weight was maximum in Hans cultivar of Phaseolus vulgaris L. -Amylase inhibitor from seeds of Hans cultivar of Phaseolus vulgaris L. was purified using ammonium sulphate precipitation, gel filtration chromatography (Sephadex G-200) and ion exchange chromatography (DEAE-Sephadex). The inhibitor was purified to homogeneity as judged by native PAGE with 34.42 fold purification and 69.35 per cent recovery. The purified inhibitor had a molecular weight of 16,218 daltons and was found to be monomer as revealed by SDS-PAGE. It was found to be heat stable upto 300C-400C and had two pH optima of 5.0 and 6.9. Nature of inhibition was found to be of non competitive type as determined by Lineweaver burk plot. The purified inhibitor was found to have inhibitory activity against -amylases extracted from larvae of Callosobruchus chinensis, Tribolium castaneum and mid gut of Corcyra cephalonica.
