The objectives was to study seminal characteristics, post thaw semen evaluation and freezability of semen, and post thaw incubation periods on frozen semen of different breeds. A total of 100 semen ejaculates from 5 breeds of bulls (20 ejaculates from each of 5 breeds) were taken during the recovery period for study. Volume, colour, consistency and concentration were recorded. A sample of fresh semen was used for evaluation of live sperm count, sperm abnormalities, acrosomal integrity and in-vitro functional tests i.e. HOST and CMPT. Fresh semen was diluted initially and a drop from it was used for evaluation of individual motility. Seminal parameters recorded during the recovery period were compared with the data available in CSS for the period of poisoning and before poisoning. However, not much data for the period of poisoning was available as all the ejaculates were discarded at that time. The volume of semen differed significantly (P≤0.01) between the breeds and was significantly lower in Murrah buffaloes during the recovery period. The volume was significantly increased in H.F. (P≤0.01), Gir (P≤0.01) and Sahiwal (P≤0.01) during the period of poisoning as compared to before poisoning and during recovery period. The colour of the semen varied from creamy to watery and consistency from thick to translucent in all the breeds during the recovery period. Curdling of semen was a major problem reported during the period of poisoning in all the breeds with the consistency varying from thin creamy to watery The concentration of spermatozoa differed significantly (P≤0.05) between the breeds during recovery period. Sperm concentration in Sahiwal breed was significantly low (P≤0.05) during the poisoning period than before poisoning and during recovery period while there was no significant difference in other breeds. The average individual motility did not differ significantly between the breeds during recovery period while it was significantly reduced (P≤0.01) in all the breeds during poisoning period than before poisoning and during recovery period. The live sperm percentage was significantly higher in Murrah and Jersey breeds (P≤0.01) while significantly lower in Sahiwal breed during recovery period. There was no significant difference in the live sperm percentage between the before-poisoning phase and the recovery phase in Murrah, H.F. and Gir bulls whereas, in Jersey bulls it was significantly higher (P≤0.05) and in Sahiwal bulls, significantly lower (P≤0.01) during recovery period than before-poisoning phase. The average total sperm abnormalities were significantly higher (P≤0.01) in Jersey, H.F. and Sahiwal breeds while lower in Murrah breed semen during recovery period. The total abnormalities were significantly higher in Murrah (P≤0.05), H.F. (P≤0.01), Gir (P≤0.05) and Sahiwal (P≤0.05) breeds during the recovery period as compared to the before poisoning period. CMPT, Percent normal acrosome, and percent HOS positive sperm did not differ significantly between the breeds during the recovery period. After 24 hrs of cryopreservation the straws were evaluated for various seminal characteristics. The percent normal acrosome and percent total sperm abnormalities did not differ significantly between breeds during the recovery period. Post thaw motility in Murrah and Sahiwal breeds was significantly low (P≤0.05) as compared to that of Jersey, H.F. and Gir bulls; The percent HOS positive sperm in frozen semen was significantly higher in Murrah bull semen (P≤0.05) and lower in Gir and Sahiwal semen; and the cervical mucus penetration distance was significantly higher in Jersey breed (P≤0.01) and lower in Gir and Sahiwal breed during the recovery period. To observe freezability of semen during the recovery period, frozen semen straws prepared from above evaluated fresh semen were observed. Freezing resulted in significant decline in post thaw motility, acrosomal integrity, CMPT, and HOST. % total abnormalities were higher in frozen semen of Murrah (P≤0.01), H.F. (P≤0.05) and Gir (P≤0.01) than the fresh semen during recovery period while it did not differ significantly in Jersey and Sahiwal breeds. Effect of post thaw incubation durations was observed by evaluating, frozen semen straws immediately after thawing at 0 hr (control) and after 15, 30, 45 and 60 min of incubation at 37°C. The overall post thaw motility in all the breeds was significantly higher in 0 min (P≤0.01) and there was a significant reduction in the motility during 15 min, 30 min, 45 min and 60 min intervals during the recovery period. The overall post thaw HOS positive sperm % and the cervical mucus penetration distance in all the breeds was significantly lower in 0 min (P≤0.01) and there was a significant increment in the motility during 15 min, 30 min, 45 min and 60 min intervals. In the present study, seminal parameters of Murrah, Jersey, H.F., Gir and Sahiwal bulls have been characterized to some extent in a selected population. Sperm concentration was significantly higher and live spermatozoa % was significantly lower in Sahiwal breed as compared to other breeds during the recovery period. Ejaculate volume was significantly higher in H.F., Gir and Sahiwal breeds during the period of poisoning as compared to before poisoning and during recovery period. Sperm motility % was significantly reduced during the period of poisoning.
