PrajapatiGawai Kunal Manohar2022-05-122022-05-122020https://krishikosh.egranth.ac.in/handle/1/5810184598The present investigation involved formulation of selective broth for coliforms and E.coli by optimizing the rate of addition of Sodium lauryl sulphate salt, Gentamicin sulphate + Amoxycillin (in 1:1 ratio) and coliform growth factor Cefsulodin by response surface methodology. From the first phase of study, formulation of selective broth for coliforms and E.coli consisting of Sodium lauryl sulphate salt, Gentamicin sulphate + Amoxycillin and Cefsulodin added at the rate of 0.25 g/100ml, 10 ul/100 ml and 312.5 ul/100 ml respectively was optimized. This combination of ingredients along with base composition of broth were able to increase the growth of coliforms. Along with this, it was able to inhibit population of Salmonella typhi ATCC 14028, Enterococcus faecalis ATCC 29212 and Staphylococcus aureus ATCC 25923 up to 2, 22 and 14 cells/ml in 10 h when spiked at 10 cells/10 ml and incubated at 37oC.EnglishDEVELOPMENT OF RAPID METHODS FOR DETECTION OF COLIFORMS IN MILKThesis
