K.2018-01-062018-01-062014-12http://krishikosh.egranth.ac.in/handle/1/5810038894An investigation was carried out to study the impact of five altered conditions representing elevated carbon dioxide, elevated temperature, elevated carbon dioxide plus elevated temperature, ambient condition and one sample from the open reference plot on the functional and genetic diversity of pink pigmented facultative methylotrophs (PPFM). The authenticity of PPFM isolates from the altered conditions was confirmed by screening for the presence of mxaF gene by amplifying a single fragment of 555 bp in all the native isolates. Based on the carbon substrate utilization test, the native PPFM isolates have been grouped under nine species viz., M. suomiense, M. populi, M. nodulans, M. aminovorans, M. mesophilicum, M. radiotolerans, M. fujisawaense, M. extorquens and Methylobacterium sp. The studies on the functional characterization of the native isolates have revealed that all the isolates were capable of producing cytokinin and IAA. Ten isolates have shown MPS activity and seven isolates were antagonistic to Sclerotium rolfsii and Fusarium oxysporum. Further, five efficient isolates based on their ability to produce IAA and cytokinin were characterised by sequence analysis of the 16S rDNA.
