Abstract
Present study was undertaken to have an in-depth study on detail epidemiological, clinico-pathological and molecular confirmation of Mycobacterium avium subsp. paratuberculosis in goats of Odisha. Goats randomly selected from ten different districts of Odisha were screened for Paratuberculosis on the basis of history, clinical signs and faecal microscopy through ZN staining. Present study recorded an overall prevalence of 38.75% with prevalence of clinical and sub-clinical Paratuberculosis were 8.06% and 30.68% respectively. Necropsy was conducted for eight goats that died during the study period. Typical clinical signs observed were frequent watery diarrhoea, pasty faeces, emaciation or lean body mass, increasingly lethargic attitude, rough body hair coat and soiled hind quarters and perineum. Present study recorded highest occurrence in Puri district (52.29%) followed by Ganjam district (34.88%) and Balasore district (45%). Chi-square analysis revealed a significant higher occurrences of disease among adult goats of more than 2-year age (51.23%), Ganjam breed (42.30%), females (39.17%), earthen floors (55.83%), rainy season (40.37%) and goats with moderate body conditions (67.5%). Present study recorded high occurrences of low shedders (n=219, 69.08%) followed by moderate shedders (n=64, 20.18%) and least numbers belonging to high shedders (n=34, 10.72%) of MAP bacilli during screening. Anaemia and leucocytosis with increased lymphocyte count was consistent in Caprine Paratuberculosis. There was significant increase in AST and ALT in Paratuberculosis affected goats. Necropsy revealed a chronic enteropathy. Histopathological lesions were mostly related to infiltrations of mononuclear cells. Molecular confirmation of MAP bacilli was done from faecal as well as tissue (intestine and MLN) samples using IS900 specific primers showing 400 bp PCR product in agarose gel electrophoresis.
