Keratoconjunctivitis Sicca (KCS) is an inflammatory disorder of the lacrimal functional unit, associated with discomfort and visual disturbance. It is multifactorial in origin and it leads to chronic ocular surface diseases, impaired vision, and a variety of complications, eventually compromising the quality of life of the animal. Current treatment modalities for KCS in dogs aims at substituting the tear film with artificial tears, stimulating tear production with lacrimo- stimulants and treating inflammation with steroidal and non-steroidal eye drops. None of the current treatment modalities focus on regeneration of functions of the tear gland. Regenerative therapy with platelet rich plasma (PRP) is reported to be promising in human dry eye patients. Considering these facts injection of autologous platelet rich plasma into the lacrimal glands, under the conjunctiva and into the third eyelid gland weekly once for four weeks was adopted in the present study. Autologous PRP was produced by double centrifugation method. After PRP therapy for one month there was considerable improvement in tear production, quality of the tear film, regeneration of ocular surface epithelium and number of goblet cells. A marked reduction in corneal oedema, pigmentation density and thereby significant improvement in corneal clarity and vision were also noticed after PRP therapy. The regeneration of lacrimal glands as well as resumption of secretions significantly improved the degree of hydration of conjunctival and corneal epithelium, the nature of ocular discharge, ocular surface health and appearance of the eye, thereby address one of the main concerns of the owners regarding dry eye. From the observations in the present study autologous PRP therapy can be advised in mild to moderate cases of KCS
