Abstract
The study was conducted to appraise the mechanisms of stripe rust resistance in four barley genotypes including three resistant (RD2900, RD2901 and RD2552) and one susceptible (Jyoti) genotypes against different races of Puccinia striiformis f. sp. hordei. Disease reaction showed RD2901 as immune to very resistant followed by RD2552 as immune to resistant and RD2900 as moderately resistant and Jyoti as susceptible against M, G, 57, Q and 24 races. RD2901 possessed higher antioxidant potential in terms of free radical scavenging activities and phenolic compounds under control conditions and maintained sufficiently high on inoculation with different races. Upregulation of NOX, POX, CAT, APX, MDHAR, DHAR, GR, PR proteins, PAL, TAL, GDH and OAT with a concomitant decrease of H2O2 and MDA in RD2901 could also play a vital role in resistance against stripe rust. It also showed maintained levels of β-glucan and increased contents of lignin, proline, glycine betaine and choline. GSH/GSSG significantly declined in Jyoti and increased or remained unaffected in resistant genotypes that might be playing a key role in imparting tolerance. 2-DE showed 36 DEPs in RD2901 under control and infected conditions. Among them, 14 proteins were over expressed and 12 proteins as newly synthesized under infected conditions. Inheritance studies showed that F2 population (RD2901  Jyoti) segregated into 31 resistant and 89 susceptible individuals that fit well to 1:3 which is expected of single recessive resistant gene. BSA indicated that Bmag0500 marker located on 6H chromosome of barley genome was found to be linked with resistant gene.
