Abstract
The present study was conducted to investigate the effects of
Melatonin (MLT) and Canthaxanthin (CX) supplementation on liquid storage
of Magra ram spermatozoa. Total 48 ejaculates were collected from eight
Magra rams (6 ejaculate from each ram) aged between 1.5 to 3 years reared
at ICAR, CSWRI-ARC, Bikaner under uniform management conditions. The
ejaculates were evaluated macroscopic for colour, consistency, total ejaculate
volume, pH and microscopic for mass motility, individual progressive motility,
sperm concentration, sperm viability and sperm abnormalities. The ejaculates
of each collection session were pooled and were split into three aliquots (C,
MLT and CX). All the aliquots were diluted (1:10) with Tris-citric egg yolk
extender (TCEY). The aliquot C was devoid of antioxidant to serve as control
whereas, the other two aliquots, MLT and CX, were supplemented with 1mM
Melatonin and 25μM canthaxanthin, respectively. Thereafter, the samples
were stored at 4oC for 72 hrs and various seminal parameters (individual
progressive motility, HOST, sperm viability, sperm abnormalities) along with
oxidative stress parameters like Total antioxidant capacity (TAOC) and
Malondialdehyde level (MDA) were quantified at 0, 24, 48 and 72 hrs of
preservation. The results indicated that the mean percentage values for
progressive sperm motility, sperm viability and TAOC were significantly higher
(p<0.05) in MLT treated group and HOST significantly higher in CX group.
Significantly lower percentage of total sperm abnormalities and MDA level
were observed in treatment groups compared to control group. It was
concluded that MLT and CX supplementation improved the liquid storage
capacity of Magra rams spermatozoa.
