VERIFICATION OF PARENTAL LINES AND HYBRIDS THROUGH MOLECULAR MARKERS IN BRINJAL (Solanum melongena L.)

Abstract
The present investigation was undertaken during Rabi-2010 and Summer-2011. The experimental materials were planted at Horticultural Research Farm, All India Coordinated Vegetable Improvement Project, Department of Genetics and Plant Breeding, Indira Gandhi Krishi Vishwavidyalaya, Raipur (C.G.). The experiment was subject to identify different parental lines; test the hybridity and to observe mean performance of parents and hybrids. The experimental materials used in the study comprised of seven parents viz., Green Long (GL), Muktakeshi (MK), Pusa Purple long (PPL), IBWL-2007-1 (IBWL), Pusa Purple Cluster (PPC), Pant Rituraj (PR), Panjab Sadabahar (PS) and their sixteen F1‟s. Two molecular markers viz., RAPD and ISSR were used to fulfill the objectives. To study the mean performance of parents and hybrids the observations were collected on days to 50% flowering, days to first picking, plant height, number of primary branches per plant, average fruit length (cm), average fruit girth (cm), average fruit weight (g), total number of fruits per plant, number of fruits per plant per picking, marketable fruit yield per plant, total fruit yield per plant and total soluble solids (%). For the parental identification and hybridity test a total of forty-nine RAPD and thirteen ISSR primers were screened. Out of which, only two RAPD primers OPA-10 and OPH-20 and only one ISSR primer i.e., UBC-808 was found to be polymorphic among the parents. For the primer OPH-20, Pusa Purple Cluster and Panjab Sadabahar showed polymorphism and for the primer OPA-10, the parent Green Long and Pusa Purple Cluster was polymorphic. A hybrid between P2 and P7 (Green Long x Panjab Sadabahar) was identified on the basis of banding pattern which resembles to female parent Green Long (P2). In another hybrid of P5 and P6 (Pusa Purple Long x Pusa Purple Cluster) banding pattern of F1 hybrid resembles to one parent Pusa Purple Long (P5). F1 hybrid of parent P6 and P1 (Pusa Purple Cluster x IBWL-2007-1), the banding pattern resembles to one parent i.e, Pusa Purple Cluster (P6). F1 hybrid, P3 and P2 (Muktakeshi x Green Long), the banding pattern found to be similar to male parent Green Long. For the primer OPH-20, an F1 hybrid between P5 and P6 (Pusa Purple Long x Pusa Purple Cluster) produced banding pattern which resembles to male parent Pusa Purple Cluster. For the ISSR primer UBC-808, the banding pattern of the hybrid (Pusa Purple Cluster x IBWL-2007-1) resembles to male parent i.e. IBWL-2007-1. In genetic diversity analysis of seven genotypes of brinjal it is found that genotype P3 (Muktakeshi) and P7 (Punjab Sadabahar) are very closely related with genetic similarity of 85 per cent and the genotype P2 (Green Long) forms a separate cluster which means that genotype P2 (Green Long) is extremely different form rest the all genotypes. The mean performance study of parents and hybrids, resulted that among the parent Green Long gave the highest yield potential, followed by IBWL and PPC (Pusa Purple Cluster). In case of hybrids it was recorded highest in GL x PPL, which is followed by GL x PS, and PPL x PPC.
