Effect of UV radiation on Beauveria bassiana (Balsamo) and its characterization

dc.description.abstractSeven Beauveria bassiana isolates viz. three procured isolates MTCC4495, MTCC 2028 and NBAIR B15, two isolates of commercial formulation (Mycojaal and Daman) and two native isolates B. bassiana F10 and B5 were evaluated for their tolerance to UV radiation, for their UV protection with different oil supplements and pathogenicity against nymphs of Lipaphis erysimi and were studied for their molecular characterization. B. bassiana commercial isolate (Mycojaal) was most UV tolerant after 60 sec of UV exposure(305 nm) with the colony forming units of 8.66×103conidia/ml and was at par with the native isolate F10 (8.00×103conidia/ml). B. bassiana conidial suspension supplemented with different oils recorded maximum CFU count (9.47×103 conidia/ml) in the soybean oil supplemented conidial suspension after 60 sec of UV exposure followed by sunflower oil, rice bran and mustard oil. The conidial suspensions of B. bassiana (109 conidia/ml) supplemented with oils were evaluated for their pathogenicity along with control (oil unsupplement) against nymphs of L. erysimi. The maximum mortality (67.18 %) was recorded in conidial suspension of B. bassiana with soybean oil which was at par with the conidial suspension supplemented with sunflower oil (66.14 %). Conidial suspension of B. bassiana F10 supplemented with oils was most pathogenic and recorded maximum mortality (82.42%) and was at par with the commercial isolate Mycojaal (82.41%) and MTCC 4495 (82.38%). However, minimum mortality 36.61 per cent was in MTCC 2028. Bioevaluation of UV exposed (60 sec) conidial suspension of B. bassiana supplemented with potential UV protectants soybean and sunflower oil against L. erysimi recorded maximum mortality in UV exposed suspension in soybean oil (63.18%) which was significantly better than sunflower oil treatment (61.67%) and control (without oil supplement) (55.55%). Fifteen fungal isolates were molecular characterized using 11 RAPD markers. A total of 76 polymorphic fragments were amplified. RAPD markers data revealed the Beauveria isolates have shown large differences in genetic similarity coefficient which ranged from 0.40 to 0.86. Highest similarity was recorded in C-1 (P Isolate) and C-2 (MTCC 6097) with similarity coefficient 0.86. Lowest similarity (40 %) was in MTCC 2028 and MTCC 4495. A single 570 bp was amplified using ITS markers.en_US
