Abstract
Survey conducted during 2009
-
2010
,
2010
-
2011
and 2011
-
2012
in the vineyards of
Jammu and Kashmir, Delhi, Maharashtra
and Karnataka
states of India showed
symptom
s
typical to leafroll disease of grapevine in
the
Nashik and Pune region
s of
Maharashtra and Delhi
. DAS
-
ELISA (double antibody sandwich
-
enzyme linked
immunosorbant assay) and RT
-
PCR (reverse transcriptase
-
polymerase chain reaction)
based tests
showed the presence of
Grapevine leafroll
-
associated virus 3
(GLRaV
-
3)
in
eight
cult
ivars
and
Grapevine leafroll
-
associated virus 1
(GLRaV
-
1)
in
two cultivars
of
samples collected from the vineyards of
Nashik a
nd Pune regions of Maharashtra. In
a
vine of
cultivar Shiraj from Nashik region
,
mixed infection of GLRaV
-
3 and GLRaV
-
1
was also f
ound.
However, both the test
s
for viruses w
ere
negative in the
t
wo leafroll
symptomatic
vines
of Delhi
.
The vineyards of
Jammu and Kashmir and Ko
ppal
district
of Karnataka
did not exhibit the symptoms of grapevine leafroll disease and serological
and PCR b
ased
assay
also confirmed the absence of this disease in these regions.
The
serology and PCR based detection method
s
are highly useful
in the indexing of
grapevine cultivars for the healthy cultivation of grapevine in the Indian viticultural
industry.
Addi
tionally, these tests can also be employed for the quarantine certification
programmes of the imported propagating materials.
