Abstract
Sugarcane (Saccharum officinarum L.) is an important cash crop widely used for sugar and bioethanol production, belongs to the family Poaceae, is well known to be cultivated in the tropical and subtropical regions worldwide. After textiles, sugar industry is the second largest agro-based industry in India that plays a vital role in the socio-economic transformation of the country. Growth rate of sugarcane crop is highly affected by the climate it prevails. It has certain stages in its life cycle, where the application of water pays large dividends. Onset of tillering, elongation of internode and grand growth phase are the most crucial stage in a crop life cycle that need adequate amount of water. Among various abiotic stress, salinity is the most severe environmental stress, impairing crop production more than 20% worldwide. Salinity induces both the ionic and osmotic stress in plant that leads to a condition of physiological drought. During present study, 10 sugarcane genotype were irrigated with normal water (control) and two different salinity level (ECiw10 dS/m and ECiw 20 dS/m) at formative stage of plant (40-150 DAP). Their morphological, physio-biochemical and molecular studies were carried out at grand growth stage (160-240 DAP), that revealed a sufficient genetic variation for salt tolerance among the10 genotypes respectively. Cluster analysis through Ward’s minimum variance based on morphological and physio-biochemical observation, these ten sugarcane genotype broadly classified in three different zones, sensitive (Co 0118, Co 0238 and CoS 8457), moderate (CoS 8279, Co 5011, CoLk 99270 and Co 5009) and tolerant ( CoPant 97222, CoS 7250, Co 98014). Pearson correlation analysis of 18 different trait shows significant correlation with each of them subsequently at different level of treatment and control condition. Moreover, molecular analysis via SSR marker categorized the genotype in three different cluster as per their amplification under PCR, while presence of six ABA dependent gene in sugarcane variety Co 5011, CoS 7250, CoPant 97222, Co 98014, CoLk 99270 confirm their ability to tolerate the salinity stress. In vitro study reveals Co 0238 to be better than Co 0118 under NaCl and mannitol stress, whereas ascorbic acid was found as a potent antioxidant for mitigating the NaCl effect. Last but not the least these investigation suggested that tolerant variety could be used as parent in breeding program.
