Abstract
With the growing preferential demands in the markets by the consumers, the food industry is altering its protein obtaining sources. Plant based protein sources are highly recommended and accepted alternatives for the consumers. Among various protein rich crops, Lathyrus can be one of its best alternatives with its high amount of protein content, easy cultivation practices, profuse yield and abundant minerals. However, Lathyrus is often criticized for its anti-nutritional components owing especially to β-ODAP (β-N-oxalyl-L-α, β-diamino propionic acid). The presence of β-ODAP in Lathyrus seeds makes it unsuitable for human consumption. β-ODAP is a non-proteinic amino acid and consumption of Lathyrus as a staple food continuously for 3-4 months can cause a neurodegenerative disease known as Neurolathyrism. Hence, the present investigation entitled- “Production and characterization of protein isolates from Lathyrus sativus and expression analysis of ODAP biosynthesis pathway related gene” was carried out to obtain low β-ODAP containing protein isolates and their characterization for stability and solubility as well as to understand the expression of gene (s) involved in β-ODAP biosynthesis under drought and cold stress conditions. The SDS profile of the Lathyrus leaf protein exhibited highly intensified protein bands of molecular weight between 29 kDa to 44.3 kDa under both the stress conditions. The comparative analysis of Lathyrus and pea protein revealed maximum protein purity at 450C temperature (75.134 % for Lathyrus and 66.53 % for pea) and pH 9.0 (61.094 % for Lathyrus and 58.973 % for pea) conditions, however, an increase in protein solubility was obtained up to pH 8.0 only. The β-ODAP content of protein isolates was found to be very low suggesting it to be an excellent component for the protein supplements. The β-ODAP gene expression analysis resulted in differential amplification of a 350 bp region with the primer PLPD1 in response to implementation of different stress conditions. The present study will provide an insight for studies on protein isolates from Lathyrus with high purity indices and to understand the genetic regulation of genes involved in β-ODAP biosynthesis pathway.
